Abstract/Summary:
Cortinarius purpurascens Fr. (Zi Mo in Chinese) which is mainly produced in the Ho-lan Mountains of Inner Mongolia Alxa League and Ningxia, is a kind of rare edible fungus. In recent years, a lot of wild fungi have been reported to have anti-tumor, antioxidant and other biological effects. So, we conducted a preliminary study on the chemical constituents of Cortinarius purpurascens, and tested its antioxidant activity and cytotoxic activity. The main contents and results are as follows:1. The Cortinarius purpurascens was extracted by methanol. Then the extract was partitioned with water and different organic solvents. Through various column chromatography and PHPLC purified, we got five compounds. Then through a variety of modern spectroscopic methods (’H-NMR,13C-NMR, HMBC, HSQC, H-H COSY, NOESY, UPLC-MS), and comparison with the reference data, the chemical structures were identified as benzoic acid, azelaic acid, oleic acid, linoleic acid and sodagnitin E. Among these compounds, sodagnitin E was isolated from this fungus for the first time.2. We selected four kinds of human cancer cell lines to assess the cytotoxic activity of the extractions and five compounds from the Cortinarius purpurascens using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method. For the proliferation of HepG2 cells, A549 cells and BGC-823 cells, the methanol extract, ethyl acetate soluble part, petroleum ether soluble part and sodagnitin E showed significant and concentration-dependent inhibitory effect; for the proliferation of Hela cells, ethyl acetate soluble part, petroleum ether soluble part and sodagnitin E showed concentration-dependent inhibition.3. We screened the DPPH radical scavenging ability of all extracts and 5 compounds from Cortinarius purpurascens through microplate reader. Wherein the methanol extract and ethyl acetate soluble part have DPPH radical scavenging activity with EC50 being 267.6 μg/mL and 145.3 μg/mL, respectively. The ethyl acetate soluble part, petroleum ether soluble part and n-butanol soluble part significantly protected ROO inducted DNA rupture in the experiment using agarose gel electrophoresis to detect the protective effect on ROO* induced DNA damage. The scavenging effects on intracellular reactive oxygen species (ROS) were evaluated using t-BOOH damaged HepG2 cells. Wherein the methanol extracts, ethyl acetate soluble part, petroleum ether soluble part and n-butanol soluble part significantly decreased reactive oxygen species production by HepG2 cells and showed protective effects on cells at a nontoxic dose.Conclusion:Cortinarius purpurascens has an inhibition effect for the proliferation of several cancer cell lines, and showed strong antioxidant capacity. The present results provided scientific data for the development of C. rufo-olivaces as functional foods with anti-cancer and anti-oxidant effects.
