Marine toxins, which are produced by algae, are enriched and accumulated in filter-feeding fish or shellfish. The contaminated seafood is preyed on by people and causes a series of poisoning accidents. It is the urgent affair that the shellfish toxin must be detected rapidly and efficiently. The thesis introduced the information about the present situations of marine pollution and the types of marine toxins. The marine toxin had five kinds such as diarrhetic shellfish poison (DSP), paralytic shellfish poisoning (PSP), neurotoxic shellfish poisoning (NSP), memory deficiency shellfish poison (ASP) and ciguatoxin (CTX). Also, the method about detection and pretreatment was introduced briefly for marine toxin detection. Meanwhile, the theory of ELIAS was elaborated. In this study, a Diarrhetic Shellfish Poisons In-Suit Pretreatment and Detection System including pretreatment and ELISA detection instruments was designed to come ture in-suit detection of DSP.The contents of this paper are as follows:1.The dissertation completed the optimization of the sample pretreatment processA sample preratment process which is suitable for enzyme-linked immune detection methods was optimized to meet the demand of the automation of shellfish samples pretreatment.Based on the national industry, standard and other sample extraction and purification methods,this process used several experiments to determine a series of pretreatment steps.2.The dissertation completed the design and test of the sample pretreatment instrumentA semi-automatic pretreatment instrument was designed to enhance the efficiency of sample pretreatment. Several modules were developed to come true the steps of pretreatment with the function of power module. And a electric circuit was established with the MSP430F149 which is a high-performance microprocessor. By the electric circuit, program of lower computer could make the modules work following time sequences given by upper computer with custom communication protocol. In the mean time, program of upper computer was designed to help people controling the flow of the pretreatment. Rcovery rate of 80μg/kg sample is 80%.3.The dissertation completed the design of the image processing algorithmAfter kit reaction,the color of the solution was determined by the concentration of toxins in sample.Based on this relationship, an algorithm was designed to analyze the concentration of toxins from the color image with image processing techniques. The algorithm included image segmentation, eigenvalue extraction and converion of color image to toxins in the sample.4.The dissertation completed the design and test of the shellfish toxins optical detection instrument based on ELISA-kit and smartphoneShellfish toxins optical detection instrument could measure concentration of shellfish poison in samples by relationship between concentration and colours. Based on the algorithm,a program was developed to achieve the detection images and compute results about the concentration. Then, a device made to weaken the interference when the program of upper computer was getting images by iPhone. And backlight of the device was also optimized to enhance qualiy of the images. Detection limit of this instrument is 0.2μg/kg. What’s more, it’s accuracy is above 97% and precision is less than 0.15.5.The dissertation completed the detection of DSP in sellfish with the Diarrhetic Shellfish Poisons In-Suit Pretreatment and Detection SystemTests were token to validate the effect of the Diarrhetic Shellfish Poisons In-Suit Pretreatment and Detection System including standard addition recovery experiment and actual sample testing experiment. All these tests showed that the design of the instrument system can meet the design requirements.
