Objective This experiment by tail vein injection of lipopolysaccharide (LPS) solution, to make the Preparation of endotoxemia myocardial injury model in rats.To investigate whether total glucosides of paeony (TGP) could play a role in myocardial protection.Methods 50 male Wistar rats were randomly divided into five groups:NS control group, LPS model group, TGP low dose group, TGP middle dose group and TGP high dose group, Each group of 10. Through the tail vein injection of lipopolysaccharide (LPS) to establish endotoxemia model of myocardial injury, the concentration of 5 mg/Kg. LPS model group and TGP intervention group injected lipopolysaccharide,NS control group injected saline volume, etc. Then low, middle and high doses of TGP were administered by gastric perfusion respectively. Histopathological changes were examined by HE staining; the contents of serum CK、CK-MB、TnI were detected by Colorimetry; the content of serum SOD were detected by Colorimetry, the content of serum MDA was detected by Total Bile Acids method; the expression of NF-κB in myocardial tissue was detected by im-munohistochemistry.Results1 Histopathology demonstrated marked inflammation in cardiac tissue with infiltration of inflammatory cells. TGP with middle and high dose alleviated inflammation in cardiac tissue.2 TGP is middle, high dose group can obviously reduce the activities of CK^ CK-MB、TnI(P<0.05).3 To the TGP after the intervention, TGP is middle, high dose group can obviously increase the activities of SOD. TGP each dose group can obviously reduce serum MDA content, there is a very significant difference compared with LPS model group significance (P <0.01)4 Expression of NF-κB was up-graded in rats with myocarditis, which was attenuated in middle and high dose-TGP groups(P<0.01).Conclusion TGP with middle or high dose can effectively inhibit inflammation in cardiac tissue of rats with endotoxin-induced myocardial injury.
