D-chiro-inositol is the mediator of insulin in intracellular regulator, which can accelerate the dephosphorylation of glycogen synthase and pyruvate dehydrogenase and increase the body’s sensitivity to insulin, reduce the incidence of insulin resistance. Chickpeas grow in Xinjiang, which seeds rich in D-pinitol and it increase by germination. D-pinitol is methylated derivatives of D-chiro-inositiol, belonging to aether compounds. D-pinitol converted into D-chiro-inositol though demethylation by acid can be reduced to a hydroxyl. This paper conducted the study on D-pinitol separation purification from Chickpeas, converted into D-chiro-inositiol and D-pinitol effect of the antihyperglycemic.The seeds of Chickpeas were germinated, which purpose was improve the amount of D-pinitol. Seven and a half days later, the amount of D-pinitol reaches the highest point, 3.05%.Determined the optimum conditions of extracting D-pinitol from Chickpeas by single factor and orthogonal experiments were:solid-liquid ratio was 1:20(g/mL) and the solution was heated for 1.5 hours at 50 ℃. In this study, a RP-HPLC method was developed to determine the amount of D-pinitol was 36.85 mg/g.201×7 and 001×4 have been used in the technological process of purifying D-pinitol. The technological conditions were:the best sample concentration was 12.00mg/mL; the best sample volume was 30.00mL; then the solution was loaded on columns at a rate of 1.0 BV/h; elution with water occured at 1.0 BV/h. Theory, we can get the quality of D-pinitol was 360mg, but we just get 318mg in fact. RP-HPLC method was developed to determine the purity of D-pinitol was 93.19%.Determined the optimum conditions of manufacture D-chiro-inositol by single factor and orthogonal experiments were:solid-liquid ratio was 1:19 (v/v) and the solution was heated for 5.40 hours at 78 ℃. RP-HPLC method was developed to determine the purity of D-chiro-inositol.The next process was directed toward treating the eluate with activated carbon, and through response surface analyzing, the optimal condition was obtained:27mg/50 mL activated carbon was added to the eluate and the mixture was stirred for 42 min at 15 ℃. The decolorization rate of DCI was 93.77%. The purity of DCI was 92.37%.It was investigated that the antihyperglycemic effect of D-pinitol on diabetes mellitus mices. Feed the mice food with high-sugar and high-fat.4 weeks later, mice injected STZ intraperitoneal and treated with D-pinitol(purity was 93.19%) from Chickpeas, which concentration was 15mg/kg、30 mg/kg、60 mg/kg.32 days later, the results as follows:1.the diabetic mice’s weight was decline, FBG, glycosylated serum protein, creatinine were significantly increased compared with the control rats. After treated with D-pinitol, the diabetic mice’s weight was increased, FBG, glycosylated serum protein, creatinine and in serum were significantly declined, so D-pinitol can control the level of diabetic mice’s FBG. 2. Treated with D-pinitol, the levels of Cr and BUN with diabetic mice’s were significantly decined compared with the control rats, so D-pinitol can improve the renal function of mice. 3. Treated with D-pinitol, the level of glucose tolerance, glycogen and AST, ALT with diabetic mice’s were significantly increased compared with the control rats. Especially, the group of 60 mg/kg BW. has the best effect, so D-pinitol can repair the liver damage and improve the ability to metabolize sugar.4. Treated with D-pinitol, the concentration of serum insulin, insulin sensitivity index and β cell function index were ameliorated significantly, so D-pinitol can improve the function of islet with diabetic mice.
