Objective:To analyze gene polymorphism of mitochondrial cytochrome C oxidase subunit 1(CO1) gene and NAD H dehydrogenase 1 (ND1) gene among strains of E.granulosus isolated from patients in Xinjiang region.To discuss the genetic variation and differences among distinct intraspecies of E.granulosus,thus, To provide a theoretical basis for the prevention and control of cystic echinococcosis in Xinjiang. Methods:Collect surgical resection specimens of HCE patients in different areas in Xiniang, in order to analyze the genetic variation of selected (CO1) gene and (ND1) gene, we used specific primers, PCR amplification, and sequencing and applied DNA software to construct a phylogenetic tree. Retrospectively analyze the epidemic history, clinical manifestations, HCE types and imaging, laboratory results, and therapeutic procedures. Results:Analysis of 52 isolates gene sequences showed that only one isolates was genotype G6, and the other was Gl. In CO1 gene, There were 10 different haplotypes,which involved 30 base mutations. In homology comparison between 10 detected haplotype sequences in Genbank,2 G1 haplotypes showed the 100%homologous sequences in their gene sequence mutations. In ND1 gene, There were 11 different haplotypes,which involved 15 base mutations.In homology comparison between 10 detected sequences in Genbank,4 Gl haplotypes showed the 100%homologous sequences in their gene sequence mutations, but others 7, without counterparts in Genbank and considered the newly discovered E. granulosus ND1 gene haplotypes. G6 gene sequence showed 100%homologous with reference sequence of complete mitochondrial genome G6. clinical manifestations different from HCE types in genotype Gl infected patients, and the clinical symptoms,HCE type similar with genotype G6. Conclusions:1. Genetic diversity was lower in E.granulosus isolates from Xinjiang, and in this study, only have found two genotypes G1 and G6.2. The gene sequences belong the same class that E.granulosus isolates and E.granulosus strains (G1), but analysis presented a comb-shaped kind in NJ tree, intraspecific mutations showed genetic polymorphism.3. Clinical manifestations different from HCE types in genotype G1 infected patients, the intraspecies variation response to dynamics, pathogenicity, antigenic antibody response, chemotherapy needs to further study.
