| Objective: To explore the level of EBNA3 A,EBNA3B,and EBNA3 C specific cytotoxic immune response in healthy volunteer,nasopharyngeal carcinoma(NPC) without treatment and nasopharyngeal carcinoma after radiotherapy using enzyme-linked immunospot assay(ELISPOT). And to provide experimental assessment for nasopharyngeal carcinoma diagosis and evaluating the change of immune status after radiotherapy. Methods: 103 healthy volunteer,62 newly diagnosed NPC patients,76 NPC6 months after radiotherapy,46 NPC 1 year after radiotherapy and 77 NPC over 1 year after radiotherapy were enrolled, all patients were pathological confirmed.The frequency and number of EBNA3 A,EBNA3B,and EBNA3 C specific cytotoxic immune response were determined using ELISPOT assay. Results:(1)The frequency of EBNA3 C specific cytotoxic immune reponse were 45.8% and 54.2% in Han ethnicity and Uyghur ethnicity respectively(P=0.035);The frequency of EBNA3 C specific cytotoxic immune reponse were20.8% and 79.2% in Differentiated type of non-keratinizing squamous carcinoma and Non-differentiated type of non-keratinizing squamous carcinoma respectively( P=0.027);The frequency of EBNA3 C specific cytotoxic immune response had no sigificant difference between other clinical features,short effects and local-relapse and metastasis group in NPC without treatment; The response cell number of EBNA3 C specific cytotoxic immune response had no sigificant difference between clinical features,short effects and local-relapse and metastasis group in NPC without treatment; The response frequency and cell number of EBNA3 A and EBNA3 B specific cytotoxic immune response had no sigificant difference between clinical features,short effects and local-relapse and metastasis group in NPC without treatment;(2) The frequency and cell number of EBNA3 A,EBNA3B,and EBNA3 C specific cytotoxic immune response had no sigificant difference between NPC and healthy volunteer;(3) The frequency and cellnumber of EBNA3 A,EBNA3B,and EBNA3 C specific cytotoxic immune response had no sigificant difference between NPC without treatment,NPC 6 months after radiotherapy,NPC 1 year after radiotherapy,NPC over 1 year after radiotherapy;Conclusions:(1) EBNA3 C specific cytotoxic immune response had differences between different pathology and ethnicity;(2) The immune response was low1 year after radiotherapy and after 1year the immune response maybe restored. |
