Objective: To study the extraction process by selecting the best extraction process Capparis; prepared by pre-experiment-free Hands Disposable Disinfectant Gels methods and seek preparation process optimization preparation Capparis gel with Hands Disposable Disinfectant gels and optimize their method of preparation is to pave the way; and establish preliminary pharmacodynamic experimental mouse model based analgesic and anti-inflammatory drug mouse models to test Capparis gels inflammatory and analgesic effect, and the establishment of adjuvant arthritis model in rats to test drugs Capparis gel anti-rheumatic effects; methods: single factor screening experiments ethanol concentration, and extraction times the amount of alcohol increase the time as a factor orthogonal experimental design; On the basis of single factor tests, with viscosity, thixotropy and yield value of overall rating as dependent variable, central composite design was adopted to optimize this gel preparation technology by the amount of carbomeer(X1,%), glycerin(X2,%) and anhydrous ethonal(X3,%) as independent variables, fitting of various mathematical equations were performed using a statistical software of Design-Expert. Preparation parameters were optimized through response surface plotted by optimum fitting equations, optimized procedure was validated through experimental preparation of Uygur medicine Capparis spinosa gels.; establish mouse acetic acid writhing test and peritoneal capillary permeability experimental adjuvant arthritis model in rats and mice preliminary study of Capparis gels wind resistance effect; results: considering finalized its extraction process was 80 ℃ water bath, plus 10 times the amount of 95% ethanol 2 times, each 0.5 hours; According to the fitting model,find the quantitative relationships between threefactors and 3 evaluation indexes;Optimal formulation was carbomeer1.5%, glycerin 10.0%, anhydrous ethonal17.5%,Press optimization presciviption conducted in verification experiment,test of the selecte optimal formulation indicated that there existed approximation between the observed and estimated vauel.; t-test analysis shows that acetic acid writhing test: Capparis gel high dose, medium and low dose group and positive group were injected acetate can significantly reduce visceral and parietal peritoneum in the abdominal cavity caused by a large area, longer inflammatory pain, wherein positive group and Capparis gels there was a significant difference between Capparis gels each dose group and positive group could inhibit capillary permeability in mice with the control group: peritoneal capillary permeability experiments; high dose group the best analgesic effect(P<0.05). And the high-dose group than the obvious, but not better than the positive group; anti-rheumatic arthritis model experiment: Capparis gel different doses significantly inhibited adjuvant arthritis model in rats compared with the model group swelling(P<0.05), have a certain impact on rat organ index, lower serum TNF-α levels(P<0.05). Conclusion: The optimum extraction by Capparis and prepared Capparis gels, According to the central composite design-response surface methodology was realized optimization of preparation process of Uygur medicine Capparis spinosa gels, the establishment of a fitting model with a good predictive.; and Capparis gels have potential anti-rheumatic effect.
