Objective: In recent years, it’s reported frequently that Polygonum multiflorum and its related preparations cause liver damage. These reports have aroused wide attention at home and abroad. It has important theoretical and practical significance to find a reasonable and effective method to take, to reduce the risk of drug use and improve the level of clinical rational drug use. This article mainly conducts the research from the different ways of taking Polygonum multiflorum, so as to provide reference for clinical rational drug use. Method: Through the establishment of Radix Polygoni Multiflori fingerprint of Polygonum multiflorum samples, which are extracted on the different preparation conditions such as different pieces of specifications, different solvents and different making wine conditions, we can carry on principal component analysis and cluster analysis, to find out the differences between different samples. We can use QAMS to determine the content of Radix Polygoni Multiflori to find out the contents of the 6 main components in different samples. By using the samples of normal human L02 hepatocytes inhibitory rate as evaluation index of liver toxicity, we can find the Polygonum multiflorum is rational and safe way of taking. Based on the spectral correlation analysis, combining chemical and biological detection technology and using methods of multivariate correlation analysis can be used to find out the toxic related compounds, to provide reference for its quality control. Results: 1: Content determination method of 6 main components of Polygonum multiflorum based on QAMS. The RSD value of each index of the system adaptability of methodology study and QAMS was less than 5%.Determination in the same time of stilbene and anthraquinones can save time and cost of experiment of each component content determination of Polygonum multiflorum. 2: Fingerprint analysis and content determination Compared with bulk pieces fingerprint powder pieces of phase had low similarity.By means of principal component analysis,bulk pieces and powdered pieces can be clearly distinguished and the dissolution rate of each component of the powder pieces is higher than that of the bulk pieces., but easy to be stuck. Compared with aqueous extract, the similarity of the fingerprints of ethanol extracts from different concentration solution were lower than those of aqueous extracts, and the contents of anthraquinones were higher than that of aqueous extract. The toxicity of ethanol extracts from different concentrations of ethanol was higher than that of aqueous extract. In the process of making wine, the fingerprints of samples without UV irradiation and with UV irradiation of different time had low similarity, and with the increasing irradiation time, similarity decreased gradually. In the process of UV irradiation, trans- THSG(2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside) THSG was converted to cis- THSG, and the conversion rate was increased with the process of UV irradiation. 3 Biological toxicity test results Powdered pieces drinks toxicity was significantly higher than that of the bulk pieces ones; toxicity of ethanol extract was significantly higher than that in aqueous extract of toxicity; Polygonum multiflorum wine with UV irradiation was more toxic than that without UV irradiation. 4 Multivariate correlation analysis Through the cell inhibition rate and the multivariate correlation analysis of six main components content, we analyzed from different pieces of sample specifications, different extraction solvents and samples,different making wine process and concluded that cis-THSG and emodin have contributed greatly to the toxicity of Polygonum multiflorum. Conclusions: 1: Determination of main ingredients of Polygonum multiflorum can simultaneously determine the content of THSG and anthraquinones. 2: Don’t take Polygonum multiflorum by making root powder,to prevent the risk of liver injury3: In the industrial production, we should take the aqueous extraction method, do not use alcohol extract, to reduce the risk of drug use. 4:Don’t take Polygonum multiflorum wine as much as possible. The making wine process must avoid light; After Polygonum multiflorum process, be dried in the shade, not in the sun.5: We can tentatively determine toxic main component of Polygonum multiflorum as cis-THSG and emodin through the analysis of the correlation...
