Background:Esophageal cancer(EC) is one of the most aggressive malignant tumors of the digestive tract in the world. The latest data show that an estimated 455800 new esophageal cancer cases and 400200 deaths occurred wordwide. Histologically, the two main types of esophageal cancer, esophageal squamous cell carcinoma(ESCC) and esophageal adenocarcinoma(EAC), exhibit different etiologic and pathologic characteristics. In China, ESCC is the predominant subtype and contributes to nearly 90% of all ECs. Although we have made some progress in the diagnosis and treatment of ESCC in recent years, but the long-term survival rate of ESCC patients had not been significantly improved due to the problems in the early detection, early treatment, prevention of recurrence and metastasis, prognostic evaluation et al, in particular, the prognostic evaluation of ESCC is still low due to the lack of effective prognosis markers. Therefore, it is significant to identify sensitive and specific biomarkers to provide valuable objective indicators for prognostic evaluation of ESCC, and to improve the level of the overall treatment of ESCC.The definition of lncRNA is an RNA molecule,who were not shorter than 200 nt that lacks protein-coding potential. Lnc RNAs play an important role in maintaining chromosome integrity, protection structure, gene imprinting, genomic transcription,translation and epigenetic regulation. More and more studies have confirmed that lnc RNAs play a vital role in tumor development.Brain Cytoplasmic RNA1(BCYRN1) is a 200 nt lnc RNA, also be called BC200, once thought to selectively expressed with primates of the nervous system, and then it is found aberrant expressed in breast cancer, lung cancer, cervical cancer, ovarian cancer and other cancer tissues. Studies have reported that c-MYC can promote the expression of BCYRN1 in lung cancer tissues and cells, thereby increase the invasiveness of cells by combining with the binding sites in the promoter region of BCYRN1. So, what is the expression of BCYRN1 in ESCC in china? and what is the relationship between the BCYRN1 expression and the prognosis of ESCC? So far there has been no relevant reports. The purpose of this study was to detect the expression of BCYRN1 in ESCC and its relationship with clinicopathological features and prognosis of patients.Objectives:To explore the expression and prognosis significance of long noncoding RNA-BCYRN1 in ESCC patients who received radical resection.Methods:Quantitative real-time polymerase chain reaction(q RT-PCR) was used to detect the expression level of BCYRN1 in cell lines EC9706, Eca-109, SGC-7901, and cancer tissue and paired adjacent normal tissue samples from 70 ESCC patients who received radical surgical resection.The correlation of the relative expression level of BCYRN1 with clinical-pathological features, and Disease Free Survival(DFS) and Overall Suvival(OS) of ESCC patients were also analyzed.Results:1.The expression of BCYRN1 in EC9706,Eca-109 and SGC-7901 was high abundance.2.BCYRN1 was upregulated in 57.1%(40/70) ESCC tissues and the relative expression of BCYRN1 was significantly higher in ESCC tissues compared with adjacent normal tissue samples(P=0.023).3.The high levels of BCYRN1 expression were not related to clinical-pathological features such as age, TNM stages, histological grade(P>0.05).4.Kaplan-Meier analysis showed high expression levels of BCYRN1 were correlated with poor prognosis in ESCC patients. Patients with a high level of BCYRN1 had a shorter DFS and OS than those with low BCYRN1 expression( x~2=4.488, P=0.034 and x~2=4.629,P=0.031).ON multivariate analysis, the HR of BCYRN1 expression was 2.165(95% CI:1.119-4.186,P=0.022)for DFS and 2.242(95%CI:1.120-4.488,P=0.023)for OS.Conclusions:Our results indicate that high expression of BCYRN1 reflect poor prognosis and might be a potential prognostic indicator for ESCC patients who received radical resection.
