Abstract/Summary:
The nearly mature fruits of Rubus chingii Hu(Rosaceae) have been used as a traditional Chinese medicine. It can benefit the kidney, reduce the frequency and semen leakage. It is one of the most significant Chinese medicines in the ancient prescription of anti-aging and prolonging life. Based on the theory of free radicals, this thesis mainly study the anti-aging effect of raspberry. The raspberry glycoprotein composition can scavenge free radicals.To apply modern theories clarify its anti-aging effect, we put the raspberry glycoprotein composition as the research object and systematically study its anti-aging activity in vivo and in vitro.The research content is mainly divided into two aspects, they are as followings:1. Researching the anti-aging effect of raspberry total glycoprotein. According to the nature of the glycoprotein, we adopt the method of water extract-alcohol precipitation combining ultrasonic assisted extraction to extract the total glycoprotein ingredients in this experiment. We can establish aging model by injecting D- galactose, view relevant literature to determine the dosage and develop a reasonable dose gradient. Weighing mice’s weight daily and observing their behavioral activity. Four weeks prior to the establishment of the aging model, then we start filling medication for four weeks, finally killed them. By comparing the differences with each group mice about their behavior, body weight, organ coefficient, liver and kidney biopsy and the antioxidant index in vivo, indicating raspberry total glycoprotein has good anti-aging activity, and there is a certain dose-effect relationship.2. Extraction, separation, purification and antioxidant activity of raspberry glycoprotein. Using DEAE- 52 and Sephadex G- 100 further separate raspberry total glycoprotein to get four kinds of single component of raspberry glycoprotein. These four raspberry glycoprotein were identified as a single component by polyacrylamide gel electrophoresis, and presumed into four homologs. Combining with technologies such as infrared spectrophotometry, we can analysis their chemical structure. Utilizing ABTS and DPPH assay method measure the four kinds of raspberry glycoprotein’s antioxidant activity in vitro. Vitamin C is a positive control, results showed raspberry glycoproteins having good oxidation resistance, which No. 3 raspberries glycoprotein showed the strongest antioxidant activity.
