Alzheimer’s disease is a common complex human neurodegenerative disease. The toxicity of pathological changes of amyloid peptide to the brain was responsible for the AD.This paper explored the mechanisms of anti-toxic effects of theaflavins extract by studying on the functions of theaflavins extract delaying the paralysis induced by Aβ in transgenic Caenorhabditis elegans Alzheimer’s disease model and the related gene expression of them.The paralysis studies have indicated that theaflavins extracts can similarly protect against paralysis of transgenic C.elegans Alzheimer’s disease model CL2006 (Aβ3-42) and GMC101 (Aβ1-42).The optimal theaflavins extract concentrations of CL2006 and GMC101 were 100mg/L and 500mg/L.At the 8th day of CL2006,compared with control group,the unparalysis rates of other groups that treated with 10mg/L,100mg/L,200mg/L and 500mg/L theaflavins extracts were improved 14.33%、21.05%.8.19% and 12.83%,the effects of repression were remarkable. And, the unparalysis rates of pretreated GMC101 groups were improved 4.92%.8.36%.9.67% and 11.97% compared to control GMC101, the protection extremely significantly.Unpretreated groups were no significant difference compare with control.By measuring the body length of C.elegans of control and theaflavins extract treated groups, there were no significant differences in them,suggesting that theaflavins extract had no harmful effects on growth and development of C.elegans.Quantitative gene expression analysis revealed that theaflavins extract repressed the expression of Aβ human both in CL2006 and GMC101.In addition,theaflavins extract up-regulated the expression of heat shock proteins HSP-16.1 and HSP-16.2 levels in CL2006.In the contrast,the expression of HSP-16.1 was repressede and the level of HSP-16.2 had no significant difference in GMC101.And the expression levels of aip-1was significant repressed in GMC101 while increased in CL2006.The levels of amy-1 were extremely significant repressed in GMC101 which treated with theaflavins and significant repressed in treated CL2006.Further studies about the mechanisms of anti-toxic effects of theaflavins extract were proceed by detecting the green fluorescent protein in CL2166 (Pgst-4::GFP) indicating that theaflavins extract up-regulate the expression of gst-4 in CL2166.
