| Objective: The aim of this study was to analised the effect of mineralization and vascularization on rabbit bone marrow mesenchymal stem cells(BMSCs). In order to research a new composite material stromal cell-derived factor(SDF1-α) loaded on Pluronic F-127 which is created by ourself. We observed the effect of this composite material by twp-part of experiment. One is in vitro, analised the effect of the composite material on the BMSCs. The other, on vivo test, in order to collect data, we placed the new composite material inside the rabbit’s maxillary sinus. The data from this study was the compared with other osteogenic materials’ effect.Methods: First, the BMSCs with 50 ng / ml SDF1-α and 0.1% w / w Pluronic F-127 complex culture. Using the thiazolyl blue tetrazolium bromide(MTT) assay testing the biocompatibility, alkaline phosphatase activity assay(AKP) for testing the substence whether the cell can vascularization, and alizarin red staining for testing the ability of mineralization. Using 18 New Zealand white rabbits outside bilaterally sinus lifting surgery, bilateral implantation of three different composites and autologous control. Three materials are: β- tricalcium phosphate, SDF-1α composite Pluronic F-127 gel and saline. After that, 18 New Zealand white rabbits were randomly divided into three groups, each group 6, respectively, after 4,8,12 w sacrificed, and X-ray observation of bilateral maxillary bone formation, HE staining, CD31, CD34, BMP-2 and VEGF immunohistochemical staining osteogenic and vascular effects, and statistical analysis.Results: MTT assay confirmed the 0.1% w / w Pluronic F-127 has no effect on cell proliferation(P> 0.05). BMSCs-SDF1α composite gel under alkaline phosphatase staining, alkaline phosphatase staining cytoplasm seen a lot of precipitation, and the remaining three groups have no precipitation. BMSCs-SDF1α composite gel, BMSCsPluronic F-127, pure BMSCs cultured in osteogenic conditions, both produce mineralized nodules, no mineralization nodule formation for non-osteogenic group. HE staining showed bone osteogenic effect of BMSCs-SDF1α composite gel and β-TCP are significant, no bone formation in saline group, in the meanwhile, BMSCs-SDF1α composite gel have significantly vascularization compared to β-TCP.Conclusion: The first time use of this new composite material in rabbit animal experiments, the SDF-1α composite Pluronic F-127 gels are still able to succeed in rabbits induced BMSCs to differentiate into bone cells, it showed non-cytotoxic and good osteogenic vascular effects. |
