Acute promylocytic leukemia(APL) is a special subtype of acute leuk emia,a malignant clonal disease.Chemotherapy is the main treatment after diagnosed with acute leukemia currently.The classical drug for APLare ATRA and As2O3. ATRA and ATO are major fusion specific APL PML/ RARαproteins, induce leukemia cell differentiation, make disease remission.Nevertheless,adverse effects of those drugs make the survival and curative of APL patients affected.These make resesrchers try to develop new drugs which have better efficacy and little adverse effects.It was reported that citral could induce some tumor cells apoptosis,and having less effect on normal spleen cells and lymphocytes.Previous study showed that citral could cause NB4 cells apoptosis,make MMP decline,the expression of Bcl-2 and NF-?B decreased,Bax gene and caspase-3 increased.We further investigated the changes of related genes when citral induced NB4 apoptosis,and provide new ideas to the development of new drugs.Objective: To investigate the mechanism of the apoptosis induced by citral in the acute promyelocytic leukemia cell line NB4Methods: NB4 cells were treated with citral for 48 h at the various concentration,and then the m RNA expression of mutation p53(mtp53) 、AIF were detected by method of q PCR.Results:(1). Ethanol group compared with the control group, the cell growth count, the expression of mtp53 gene and AIF gene were no significant different.(2).Each citral experimental group compared with the controlgroup,thecellgrowthcountdecreased,thedifferencewasstatisticallysignificant.citralbetweenexperimentalgroupswasstatisticallysignificantinthecellcount.(3).eachcitralexperimentalgroupcomparedwiththecontrolgroup,theexpressionofmtp53 mrnaaredecreased,withcitralcon-centrationincreasing(5,10,20μg/ml),mtp53 mrnaexpressiondecreasedgradually.(4).eachcitralexperimentalgroupcomparedwiththecontrolgroup,theexpressionofaifmrnaareincreased,withcitralcon-centrationincreasing(5,10,20μg/ml),aifmrnaexpressionincreasedgradually.conclusioncitralcaninduceapoptosisinnb4cellsbydownregulatingtheexpressionofmtp53 andupregulatingtheexpressionofaif.
