Xinjiang Uygur, Han Rheumatoid Arthritis Gene Polymorphism Studies

Objective: To analyze gene polymorph of STAT4( rs7574865), TAGAP(rs1738074), CCR6( rs3093023) and their relationship with ethnic Han Chinese and Uighurs in Xinjiang,and explore whether these three genes are susceptibility genes. Methods:200 patients with RA(Han, Uygur each 100),200 healthy people(Han, Uygur each 100) were did by case-control study. By the method of PCR-RFLP- sequencing all genes studied non-coding regions of the three candidate sites genotyping assay. On the test results compare allele and genotype frequencies, paired linkage disequilibrium(LD) and haplotype construction, analyze the relationship between the various polymorphisms and haplotypes and RA and its subtypes. Result: 1 STAT4( rs7574865) gene polymorph and RA were found as follows: compared with Uighurs who were healthy, Uighurs control group, a statistically significant difference(P<0.05, OR=0.641, 95%CI=0.423~0.971). It is statistics significance at difference of Han RA and Uighurs RA(P<0.05, Cp=0.180). Significant difference of GG and GT gentoype frequency between the two groups of patients with statistical significance(P<0.05, OR=0.452, 95%CI=0.239~0.852). 2 Study TAGAP rs1738074 polymorphism found: Allele frequencies were compared between the two groups and allele frequencies, the difference was not statistically significant(P>0.05). TAGAP(rs1738074) gene polymorph and RA were found as follows: All the gentoype frequency and allel frequency were compared in pairs, difference among these groups had not statistical significance(P>0.05). 3 CCR6(rs3093023) gene polymorph were found like these: Making a Comparison of allele frequencies and allele frequencies between Uighurs RA and Uighur normal control group, the difference was statistically significant(P<0.05, Cp=0.182), G, T allele frequency may lead to the pathogenesis of Uighur RA(OR=0.641, 95%CI=0.423~0.971), the diference between GA and AA gentoype frequency were statistically significant(P<0.05, OR=2.983, 95%CI=1.201~7.407),the diference between GG and AA were statistically significant(P<0.05, OR= 3.150, 95% CI=1.243~7.981). Conclusion: rs7574865(STAT4) may be the allele who cause to Xinjiang Uygur RA, These data suggest that rs7574865(STAT4) is a gene loci that lead the different incidence rate of RA between Uygur and Han. rs1738074(TAGAP) polymorphism and Xinjiang Uygur, Han RA susceptibility has no significant correlation, but does not exclude the other gene polymorphic loci is associated with RA. rs3093023(CCR6) may be associated with Xinjiang Uygur RA susceptibility.
