Objective: To study the effect of T cell factor in ADAM33 gene expression on airway wall vascular smooth muscle cells. Methods: Human airway wall vascular smooth muscle cells were cultured and stimulated with different concentration T cell factor(IL-4、IL-13、IFN-γ),The expression of ADAM33 mRNA was evaluated by RT-PCR and protein was evaluated by Western blot in different stimulated time and concentration, to make sure the relationship between ADAM33 gene expression on airway wall vascular smooth muscle cells and IL-4、IL-13、IFN-γ. Results: ADAM33 was expressed in airway wall vascular smooth muscle cells. IL-4、IL-13 enhanced ADAM33 gene expression in airway wall vascular smooth muscle cells in a dose- and time-dependent manner, while IFN-γdecreased ADAM33 gene expression. When we do the time-dependent test, the concentration of IL-4、IL-13、IFN-γ was 100ng/ml, IL-4、IL-13 enhanced ADAM33 gene expression, while IFN-γ decreased ADAM33 gene expression, after 48 h, ADAM33 gene expression go a little higher than before. Conclusions: These results suggest that ADAM33 gene is expressed on airway wall vascular smooth muscle cells and IL-4、IL-13 can enhance ADAM33 expression,while IFN-γ can decrease ADAM33 gene expression.
