| Malignant tumors are the first killer of human, which is generated from non-regulatory cell proliferation. At present, ML (malignant lymphoma) is one of the fastest rising morbidity malignant tumors in China, and it has ranked the top ten malignant tumors. Malignant lymphoma is a kind of malignant tumors which comes from hematopoietic and lymphoid tissue, and which’s pathogenetic locations are always immune cells of lymph nodes and (or) lymphoid tissue outside lymph nodes. There exists different clinical manifestations, pathological types, the response to therapies and prognosis in different types of lymphoma.ML can fall into two broad categories based on pattern of onset, lymph node involvement, illness progresses:Hodgkin lymphoma (HL) and non-Hodgkin lymphoma (NHL). HL is characterized by Reed-Sternberg (RS) cells and it contains T-cell lymphoma and B-cell lymphoma. At present, malignant lymphoma is diagnosed according to clinical histopathology, while it can not reflect the condition of clinical stages, infiltration degree, therapy and prognosis. Therefore, in addition to clinical histopathological diagnosis and imageological examination, we found that some of the other diagnostic indicators is more and more important.Objective:We screened TCP-1 protein by cell transfection, two dimensional gel electrophoresis and mass spectrometry, which may be associated with the expression of P2X7 receptor, so as to have a further study on P2X7 receptor’s function and features. And then, we detected the expression of P2X7R and TCP-1 by immunehistochemistry, and calculated the positive expression rate of these two molecules respectively for further study in the relationship between these two molecules and lymphoma. In order to investigate the relationship between P2X7R receptor and TCP-1 further, we had observation of P2X7 receptor and TCP-1’s expression in the same one lymphoma pathologic slice to show the correlation between them. And it will be a basis of the study to prove these two molecules has a close relation with lymphoma disease.Method:To study the correlation of P2X7 receptor and malignant lymphoma. First, we injected P388D1 lymphoma cells which are transfected by P2X7RshRNA and Control shRNA to foot pads of DBA/2 murines. And then, when it occurred lymphoma metastasis,we screened TCP-1 which may be related to P2X7 receptor’s expression in murine’s transferred lymph node by two-dimensional gelelectro-phoresis and mass spectrometry and to analysis TCP-1’s protein level by Western blotting and Immunohistochemistry with lymph glands(The data was not shown). Then, in order to prove these two molecules to be related to lymphoma disease, we detected P2X7R and TCP-1 in the 72 cases of lymphoma and 17 cases of control group (chronic inflammation and reactive hyperplasia of lymph nodes) pathologic slices by immunohistochemistry. After that, we observed the expression of P2X7R and TCP-1 in lymphoma pathologic slices, and expounded probable correlation with the occurrence and development of lymphoma. We observed the expression of P2X7R and TCP-1 in the same lymphoma group and control group pathologic slices, and counted the results of immunohistochemistry. In addition to, the statistical method used for analysis was exact probabilities in fourfold table to verify the correlation of these two molecules in the lymphoma disease.Results:Detecting P2X7R and TCP-1 in the lymphoma pathologic slice by Immunohistochemistry, we found that P2X7 receptor expressed on the surface of lymphoma cell membranes and TCP-1 expressed in the cytoplasm of lymphoma cell. We calculated the percentage of positive cells and did some statistical analysis of positive results.The result is that P2X7R and TCP-1 had a high positive rate in lymphoma disease and had a low positive rate in control group. And also we found a positive correlation between these two molecules. Results statistically analysis showed that there was positive correlation between P2X7R and TCP-1. The statistical method used for analysis was exact probabilities and the results between these two groups existed statistically significant difference (P<0.05).Conclusion:Based on our study, detecting P2X7R and TCP-1 in the lymphoma disease and control group pathologic slices by immunohistochemistry, the result is that P2X7R and TCP-1 have a higher positive rate in lymphoma disease, and these two molecules have positive correlation. It is indicated that the conclusion may provide a new research leads in combination diagnosis and clinic treatment. Meanwhile, it may lay a foundation for researching the relationship between these two molecules and other tumors. |
