Objective:Toexplore the influences of lung cancer cells secrete in tumor growth. From the angle of molecular biology to elucidate Andrographolideinterfere with lung cancer cells secrete STAT3 proteins to influence the growth of tumor cells.Method:Different lung cancer cells in vitro culture,the different expressions of STAT3 protein in different lung cancer cell lines were analyzed by Western blot.EGF regulate on the STAT3 protein secretion of A549 cells and H1299 cells by Western blot. The comparison of A549 cells and H1299 cells transfection efficiency by fluorescence microscope.39 nude mice without thymus were randomly divided into 3 groups, namely (1) blank serum group:0.2ml physiological saline/one nude mice; (2) A549 cells group:0.2ml physiological salinewith1×107/ml A549 cells/one nude mice; (3) H1299 cells group:0.2ml physiological salinewith 1×107/mlH1299 cells/one nude mice, After 30 days, take photos of tumor in vitro.Andrographolide regulate on the STAT3 protein secretion of H1299 cells by Western blot.Result:Westernblot can be used to determine the expression of different lung cancer cells, which were when A549 cells. H1299 cells. H460 cells. HCC-827 cells. CRL-5844 cells. PC-9 cellsis consistent, A549 cells secret STAT3 protein less then others, H1299 cells secret STAT3 protein more then others. We choose A549 cells and H1299 cells. EGF promote the secret of STAT3 protein from H1299 cells. H1299 cells transfected into P53 cannot secret STAT3 protein.Fluorescence microscope to detect the transfection efficiency of H1299 cell is relatively higher than the transfection efficiency of A549 cells. Nude mice tumor experiment results show that tumor of H1299 cells group is bigger then A549 cells group, while A549 cells and H1299 cells group is bigger than that both and tumor size of this group is relatively stable. Andrographolide promote H1299 cells and H1299 cells transfected into P53 the secret of STAT3 protein.Conclusion:STAT3 protein secretion of Tumor cells affects the size of the tumor mass formation of H1299 cells and A549 cells, tumor H1299 cells secreted strong STAT3 proteins which make nude mice tumor bigger; Andrographolide promote the secretion of STAT3 proteins of tumor cells, but for H1299 cells nude mice tumor inhibiting or promoting effect is not clear, this topic is not discussed, it is need to further research.
