Objective:To investigate the influence of Portulaca extracts on Tumor Necrosis Factor-α (TNF-α), Interleukin-4(IL-4), Histamine H1(HRH1) and Interleukin-31(IL-31) in skins of rats with atopic eczema(AE). To study its efficacy and possible mechanism in the treatment of acute eczama.Method:40 SD rats were randomly divided into normal control group, model group, Cortex Phellodendri group and Porulaca group. Except the normal group, the hair of rat’s abdomen were shaved by electric electric shaver, with an area of about 2 cm X 2 cm. Then,2,4-dinitrochlorobenzene was used to make rats acur first sensitization. After 3 days,2,4-dinitrochlorobenzene was used again on the right back of rat, the area is 2 cm X 2cm, the left back as the control site. To use excitation on rat every 3 days,3 times in a row.On the 10th day, began to treatment, two times a day, each time in the morning and the afternoon, nine days in a row.Model group, Cortex Phellodendri group and Porulaca group, respectively, give 1ml sterile distilled water, fluid of compound cortex phellodendri and purslane extract.21 days later, all animals were sacrificed, two pieces of eczema skins were taken out, The first piece of skin, placed in 4% paraformaldehyde solution fixed 48 hours, to do histopathological examination. The second piece of skin, placed in 10% neutral formalin fixed 48 hours, to do immunohistochemical examination.Results:1.The results of macroscopic observation of rat skinAt the 9th day, after 2,4-dinitrochlorobenzene was used, the right back of rats in model group appeared obvious erythema, erosion and leakage, which meant the modeling succeeded. After medication, The symptoms of Cortex Phellodendri group and Purslane group were significantly reduced. The improvement of symptom in Purslane group is more apparent.2.The results of histopathological examination of rat skinThe pathological grade of model group was 2.20±0.63. Compared with the normal group, the pathological grade of model group was significantly increased (P<0.01). The pathological grade of Cortex Phellodendri group and Purslane group were 0.70±0.82 and 0.40±0.70. Compared with model group, the pathological grade of Cortex Phellodendri group and Purslane group were significantly decreased(P<0.01). Compared with positive drug group, Purslane group were a little decreased(P<0.05).3.The results of immunohistochemical examination of rat skinCompared with the normal group, the levels of TNF-α, IL-4, HRH1 and IL-31 of model group was significantly increased (P<0.01). Compared with model group, the levels of TNF-α, IL-4, HRH1 and IL-31 in rats skins of Cortex Phellodendri group and Purslane group were significantly decreased(P<0.01). Compared with positive drug group, the levels of TNF-α, IL-4, HRH1 and IL-31 in rats skins of Purslane group were a little decreased(P<0.05).Conclusion:Purslane extracts has obvious inhibitory effect in atopic eczema, it can reduce the pathological damage, suppress the vasodilation in inflammation parts, reduce the permeability of blood vessel walls and control the leukocyte chemotaxis, its possible mechanism is by reducing the proinflammatory factor, and moderating the anti-inflammatory factor and the ratio between them. At the same time, Purslane extracts can relieve itching symptom,its possible mechanism is by reducing the itching factor,...
