Objective:Establishment of insulin resistant rats by feeding with high fat and high sugar diet, and investigate the influence of electroacupuncture on the fasting blood glucose(FBG), the plasma insulin in rats(FINS), and the expression of Ras, ERK, JNK and p38MAPK protein in the skeletal muscle of insulin resistant rats. Discuss the effect of electroacupuncture on the Ras-MAPK insulin signal transduction pathway of insulin resistant rats, and provid experimental basis of acupuncture can improve insulin resistant.Methods:The 40 male SD rats were randomly divided into blank and model group. The blank group was feeding ordinary diet, the other group was feeding high fat and high sugar diet to build the insulin resistant rats. The fasting blood glucose was determinated with glucose oxidase method, the plasma insulin in rats was determinated with radioimmunoassay, by the time calculate the insulin sensitivity index (ISI)and insulin resistant index(Homa-IR). After determining the successful modeling, the model group was divided into model 1, model 2, EA1 and EA2 groups. Each group have 8 rats. The EA1 and EA2 group were treated with electroacupuncture at Zusanli, Sanyinjiao and Weiwanxiashu,20min each time, once a day, treatment for 14 consecutive days. After all the treatment, using the Western blot detecting the expression of Ras, ERK, JNK and p38MAPK protein in the skeletal muscle of rats, and compare the change of FBG, FINS, ISI, Homa-IR, Ras, ERK, JNK and p38MAPK.Result:1.The high fat and high sugar diet Successfully established the insulin resistant rats.2.Compare with the model 1 and model 2, the FBG and FINS of EA1 and EA2 was significantly decreased, the difference have statistical significant.3.Compare with the model 1 and model 2, the ISI of EA1 and EA2 was significantly higher, the Homa-IR was significantly decreased, showed that the insulin resistance status was improved.4.Compare with the model 1 and model 2, the expression of Ras and p38MAPK protein of EA1 and EA2 was significantly higher, the difference have statistical significant.5.Compare with the model 1 and model 2, the expression of ERK and JNK protein of EA1 and EA2 was significantly decreased, the difference have statistical significant.Conclusion:1.Electroacupuncture can reduce the FBG and FINS of the rats were feed by high fat and high sugar, and improve insulin resistant.2.Electroacupuncture can increase the expression levels of Ras and p38MAPK protein, and improve insulin resistant by regulating Ras-MAPK insulin signal transduction pathway.3.Electroacupuncture can reduce the expression of ERK and JNK protein, and the effect of PI3K and Ras-MAPK insulin signal transduction pathway is negative feedback, to improve insulin resistant.
