Study On Functional And Toxical Effects Of Purple Dendrobium And Ginseng Tablets

OBJECTIVE To evaluate the function and toxicological security of purple dendrobium and ginseng tablets.METHODS Experiments from function, genetic toxicity(Ames test,micronucleus test of bone marrow in mice,sperm shape abnormality test in mice), acute toxicity and thirty-day feeding.The way of administration is based on "health food inspection and evaluation of technical specifications"(2003 version).Observing the situation of administered group and the the control group,so that we can make records.RESULTS In function test, T lymphocyte transformations capacity induced by Con A shows high dose group was higher than the negative control group of the optical density value(P<0.01).Compared with the negative control group, high and middle dose groups could significantly increased the function of DTH induced by SRBC(P<0.05).The above description show the results of cellular immune function were positive. The result of humoral immunity assay were positive, because of comparing with the negative control group, the high dose group could increase the levle of serum hemolysin(P<0.05).The carbon clear-ance method means high and middle dose groups’ phagocytic index were higher than the negative control group(P<0.01 or P<0.05).So we can draw a conclusion about the monocyte- macrophage function assay, the results were positive. All of the three groups could raise the phagocytosis rate and phagocytic index compared with the negative control group(P<0.01). High and middle dose groups’ NK cell activity were higher than control group(P<0.01).The above description show the results of NK cell activity assay were positive.In acute toxicity test and thirty-day feeding test,there was no symptoms of poisoning or death in both rats and mice,and body weight were all increased.So we can see the oral maximum tolerated dose was 16.0g/kg for rats and mice.About the thirty-day feeding test,the result of body weight and weekly intake of rats was no significant difference to the normal control group(P>0.05). Female rats’ HGB value of the high dose group was higher than the normal control group(P<0.01). Male rats’ BUN value of the low dose group was lower than the normal control group(P<0.01 or P<0.05). Male rats’ ALB value of the low dose group was lower than the normal control group(P<0.01). There was no significant difference between the remaining dose group and control group(P>0.05). And no significant external appearance and pathological changes of liver, spleen, kidneys and gonads(testes/ovaries) were discovered(P>0.05). There was no dramatically toxic effects on routine blood,biochemical index and organ ratio changes in rats,and no significant appearance and tissue sections’ changes of liver, spleen, kidneys and gonads(testes/ovaries) were discovered.So we can judgment the resultsof thirty-day feeding test was negative.In genetic toxicity test,all of the three groups’ micronucleus rates were nearly to the negative control group(P>0.05). But there was a significant difference between the negative control group and positive control group(P<0.01).So the result of the micronucleus test of bone marrow in mice was negative.There was a significant difference between the negative control group and positive control group(P<0.01), however all of the three groups’ sperm deformity rates were similar to the negative control group(P>0.05).Then the result of the sperm shape abnormality test in mice was negative.The result of Ames test was also negative.In summary,the result of the genetic toxicity test about the purple dendrobium and ginseng tablets was negative.CONCLUSION The way of administration is based on "health food inspection and evaluation of technical specifications"(2003 version), according to the the results of functional experiments, the purple dendrobium and ginseng tablets does has enhanced immune function, and results of the remaining tests were all negative,which show that the purple dendrobium and ginseng tablets is safe and non-toxic healthy food.
