Objective:P53 gene is found that the highest correlation with human tumor gene. Serum P53 autoantibodies is the body’s immune response to the mutant P53 protein product. This research adopts a new Polypeptide Microarray technology to explore glioma patients serum P53 autoantibodies expression and correlation of clinical malignant degree.Methods:In 48 cases of human glioma tumor tissue and serum, and 20 cases of normal human serum for the study, peptide microarray technology were used to detect P53 autoantibodies in response to the polypeptides, polymerase chain reaction(PCR) detection in the organization P53 mRNA expression, Immunohistochemistry(IHC) method to detect the expression of P53 protein in organization.Results:48 cases of gliomas in serum samples, 13 patients with grade I- II gliomas, among them of P53 autoantibodies high responders 4 cases accounted for 30.8%;Grade III gliomas 24 cases, of which the P53 autoantibodies high responders 10 cases, accounted for 41.7%;Grade IV gliomas in 11 cases, including the P53 autoantibodies high responders in 1 case, 8.3%;Of 20 patients with normal P53 autoantibodies high responders in 1 case, accounting for 5%. Different grades of glioma and normal person have obvious difference. P53 autoantibodies and P53 mRNA level and P53 protein expression in immunohistochemical no obvious correlation. To follow-up of 48 cases of glioma, among them 4 cases were lost to follow-up, 14 deaths, the shortest follow-up time in March, the longest follow-up of 34 months. P53 autoantibodies in patients with high expression of lower expression of patients with longer survival time, Which has significantly statistical differences.Conclusions:Preliminarily concluded that the ratio of normal serum P53 autoantibodies is less. Glioma patients serum P53 autoantibodies response significantly, of which the P53-8 is the most common, and show the higher SNR.Peptide microarray technology to detect serum P53 autoantibodies in predicting clinical malignant degree and prognosis of glioma patient have certain effect.Convenient and quick detection in patients with serum P53 autoantibodies can dynamically test many times.This found has great value in postoperative follow-up and prognosis judgement of glioma patient.
