Association Between Height-related Genes SNPs And Adult Height Of Han Population In Guizhou Province

Objective:Human height is influenced by many factors, it is a classic quantitative trait. The height of an individual is the result of many growth and development processes, heritability of height was about 80%, and the inheritance pattern of stature can be explained by the combination of many gene effects. Nowadays, hundreds of candidate genes have been reported associated with height. It was reported that single nucleotide polymorphisms could explain height variation, they may influence the individual’s height variation through combined effects. Many studies reported the correlation between SNPs and adult height. Here, we study the correlation between the polymorphism of HMGA2,GDF5,NPR2,ZBTB38,LIN28B,GPR126 and the onset of height, we analyzed differences of genotypes frequencies of thirty SNPs of Han population in Guizhou province.And we also analyzed the possible mechanism of the differences of height among different genotypes of each SNP.Methods:1069 healthy Hans (364 males and 705 females), who were long-term residents of Guizhou Province, were recruited. Physical examination and questionnaire were carried out, and the genomic DNA was extracted. SNPs were determined by MS MALDI-TOF. We verified the 30 SNP loci in the Chinese Han nationality using MassARRAY Sequenom system. Association analyses were performed using one-way ANOVA and linear regression analysis.Results:1. Men’s and women’s height were normally distributed(p>0.05). The average height of male is 165.55 cm, and 153.64 cm of female.2. The genotypes of HMGA2at rs1351394, rs7968682, rs8756 sites were associated with male height. At rs1351394 site, men with the CT genotype were 2.03cm (p=0.016) taller than those with CC genotype. At rs7968682 site, men with the GT genotype were 2.41cm (p=0.006) taller than those with TT genotype. At rs8756 site, men with the CA genotype were 2.40cm (p=0.006) taller than those with AA genotype.3. The genotypes of GDF5 at rs143383, rs143384, rs224331, rs6060369 sites were associated with female height, each explained 1.4%,0.9%,1.1%,1.0% of total variance in height (P=0.026,0.021,0.023 and 0.022). At rs143383 and rs143384 sites, women with the GG genotype were 1.67 cm (P=0.002),2.29cm (P=0.024) and 1.55cm (P =0.036),2.13cm (P=0.004) taller than those with AG and AA genotypes. At rs6060369 site, women with the CC genotype were 1.74cm (P=0.024) and 2.21cm (P=0.004) taller than those with CT and TT genotypes.Comclusions:The polymorphisms of HMGA2,GDF5,NPR2,ZBTB38,LIN28B,GPR126 genes existed in Chinese Han,and the alleles and genotypes frequencies distribution conformed to Hardy-Weinberg Equilibrium. The polymorphisms of HMGA2 gene(rs1351394,rs7968682,rs8756)might be associated with male height, and the polymorphisms of GDF5 gene(rs1043383,rs143384,rs224331,rs6060369)with female height. They may be ones of the genes which affect adult height in Chinese Han population. Our findings increase the number of genetic factors of height, suggest additional genetic factors that may contribute to its individual difference.
