| Objective: To study the effect of P90 ribosomal S6 protein kinase 2(RSK2) in survival of HeLa cells in hypoxia and its mechanism.Methods: To study the regulatory role of RSK2 in glycolysis and the effect RSK2-mediated glycolysis on adaptive survival of HeLa cells in hypoxia. Cell biology, molecular biology, and pharmacology methods were used in this study.Results: 1) In hypoxia, RSK2 contributes to HeLa cells survival in hypoxia, as evidenced by the increases in the ability of proliferation and colony formation, and decreases in the amount of cleaved caspase-3 and PARP. 2) Furthermore, we found that RSK2 acts as a novel regulator of glycolysis in hypoxia, as evidenced by the increases in glucose consumption, lactate production and glycolysis-mediated ATP supplement.Conclusion: Glycolysis activation induced by RSK2, plays an important role in enhancing HeLa cell survival in hypoxia. This finding should help further understand the molecular and cellular functions of RSK2 in cancer, and may provide a potential target for cancer prevention and treatment. |
