Evaluation Of Left Ventricular Function In Swine Model With Myocardial Infarction After Being Implanted Induced Pluripotent Stem Cells Using Dual-source CT

Objective:To investigate the influence of being implanted induced pluripotent stem cells(iPSc) on the left ventricular(LV) functional change and ventricular remodeling of myocardial infarction( MI) swine model by using dual source CT-coronary angiography(DSCT-CA). Methods:Eighteen swine were randomly equally divided into control group,MI group and iPSc group. The LV systolic function and movement of all groups were assessed with dual source CT at the 1st and the 6th week after operation respectively, including the left ventricular end—systolic volume(LVESV), left ventricular end—diastolic volume(LVEDV) and left ventricular ejection fraction(LVEF). The results were compared in each group and among the three groups. Surviving iPSc cells in infarcted myocardium were identified by triphenyltetrazolium chloride(TTC) stain and hematoxylin and eosin(HE) stain 6 weeks after the transplantation of iPSc. Results:Comparing with the control group, LVEF decreased(P<0.0001) in the MI group and iPSc group at the first week after operation. Six weeks later, the MI group showed increased LVESV(P=0.010) and decreased LVEF(P<0.001) in relative to the control group. The iPSc group showed increased LVEF(P=0.003) compared with the MI group. LVESV and LVEDV increased(t=3.86,P=0.012;t=2.96,P=0.031) in the MI group at the sixth week after operation compared with the first week’s result but there was no significant difference with LVEF. The iPSc group showed increased LVEF(t=3.14,P=0.026), but the LVESV showed no significant difference between the first and sixth week after operation.HE staining and TTC staining showed there was large infarction area, and no viable myocardium cells in the MI group. Compared with MI group, there were less infarction area and more viable myocardium islands in the iPSc group. Conclusions:1. Implantation of iPSc into the infracted myocardium might decrease theinfarction area and improve LV systolic function and movement 6 weeks later.2. DSCT-CA is useful in evaluating the success of MI model and LV remodelingand systolic function after being implanted iPSc to estimate the prognosis ofclinical treatment in acute myocardial infarction.
