Abstract/Summary:
Alcoholic liver disease is the second largest category of liver disease after infectious liver disease. Excessive alcohol consumption on the health of the threat and social harm, is the world’s important public health issues. This topic through the study the impact of different factors of Oolong tea polysaccharides extraction rate and obtain the optimum extraction process, and through the construction of acute alcoholic liver injury in animal models, of Oolong Tea Polysaccharide on acute alcoholic liver injury in mice protective effect. And provide a certain basis for the evaluation of the effectiveness of its health care.This article examines the use of single factor analysis of solid to liquid ratio, extraction time, extraction temperature, ultrasonic power extraction process of Oolong tea, and to determine the optimal conditions for using response surface analysis. Then as alcohol induced acute liver injury model in mice was established, of Oolong Tea Polysaccharide on mice animal model of behavior, survival rate, body weight, liver coefficient and the tolerance time and the time of drunkenness, and SOD, MDA, GSH-PX in mice serum ALT, AST and liver tissue.Through the use of alcohol induced rat model of alcoholic liver injury. The results showed that:compared with model group, treatment group mice, mice after intragastric administration of alcohol tolerance time have different levels of variable length, in the mice drunken sleeping time also reduced in different degree; oolong tea polysaccharides will contribute to elevated activity of GSH-PX in the liver of mice, have certain inhibition to raise the content of MDA in liver tissue; from the liver staining histological morphology of display more fatty liver cell vacuoles and inflammatory cell infiltration in the model group, the liver, liver injury significantly. The liver regeneration coefficient of the drug group was significantly increased, and the alcohol induced liver injury was reduced to different degrees.The results of response surface optimization show that the extraction power results show that the extraction power, extraction time and extraction temperature have a significant effect on the yield of Oolong tea polysaccharides. By ultrasonic assisted extraction conditions of Oolong tea polysaccharide, extraction power was 300W, extraction power was, extraction time was 41min and extraction temperature was 82, the extraction rate was 4.07mg/g, and the error of the model was 3.55%. The results showed that the model was reasonable and consistent with the model. Compared with the traditional extraction method, the average yield is 4.07mg/g, the extraction effect is good, the ultrasonic assisted extraction can greatly save the extraction time, reduce the extraction cost, and the polysaccharide yield is also higher.General speaking, oolong tea polysaccharide on alcohol induced liver injury has a protective effect, which to oolong tea polysaccharide in the dose group has the best effect, which may be related to reduce in mice serum levels of free radicals, inhibition of lipid per-oxidation. The results of this study provide a theoretical basis and technical support for the prevention and treatment of alcoholic liver injury, and to provide theoretical basis and technical support for the development of Oolong tea polysaccharides and the development of compound nutrition and health care products.
