Background:Tanshinone ⅡA is a kind of fat soluble component extracted from Salvia miltiorrhiza. It has a wide range of pharmacological effects, small side effects, low cost, etc. The results showed that the expression of Tanshinone IIA could not only regulate the expression of CYP11B1 and CYP11B2, but also inhibit the synthesis of cardiac aldosterone. It can inhibit the expression of P- and ICAM-1 in early inflammatory reaction, and reduce the inflammatory reaction in ischemic reperfusion injury.HMGB1 is a newly discovered important "late" inflammatory cytokines, and not only can promote TNF-a, IL-1, ERK kinase, P38MAPK kinase, JNK kinase, and promote nuclear transcription factor NF-nuclear translocation B. These studies suggest that HMGB1 may be a central link in the regulation of inflammatory cytokines, not only the biological effect of its own secretion, but also regulate the secretion of other inflammatory factors. It has been widely studied and demonstrated in the liver, spinal cord, brain and other tissues.The aim of this study was to investigate the effect of different doses of IIA on the expression of HMGB1 in the model of myocardial ischemia reperfusion injury, in order to explore the possible mechanism of its protective effect on myocardial ischemia reperfusion injury.Method:The myocardial I/R injury model in rats was established with Normal group, IR group and low dose TSA-treated IR group (L-TSA), middle dose TSA-treated IR group (M-TSA) and high dose TSA-treated IR group (H-TSA). Myocardial pathological change were detected by HE staining, the expression of HMGB1、SOD、MDA in Plasma were detected by ELISA. the expression of HMGB1、TNF-α mRNA and protein in myocardium were detected by PT-PCR and Western blots.Results:Compared to IR group, The myocardial pathological changes after acupuncture preconditioning were reduced by HE, and the expression of plasma HMGBlwere decreased, and the expression of myocardial HMGB1, TNF-α were also decreased in a dose-dependent manner. Conclusions:Tanshinone IIA pretreatment may play a protective effect on myocardial ischemia reperfusion injury, the mechanism may be related to the lessen of HMGB1 mediated inflammatory response.
