Objective:To investigate the effect of dexmedetomidine on spinal cord ischemia-reperfusion injury in rats, and to explore its possible mechanism.Methods:96 adult SD rats were divided into four groups (n= 24) by random number table method:sham operation group (S), ischemia-reperfusion group (IR), dexmedetomidine treatment group (DEX),combination treatment of dexmedetomidine and yohimbine group (DY), each group was divided into four subgroups (n= 6) according to the time point after ischemia-reperfusion injury (12,24,48 and 72 hours). After the spinal cord model was established by the improved Zivin method,The moting function of hindlimbs was evaluated by BBB scale, L4.6 spinal cord pathological changes were observed by HE staining,the expression of Bcl-2 and Bax was examined by immunohistochemical,staining,apoptosis rate of cells was calculated by TUNEL, and the expression of caspase3 was detected by Western bolt.Results:Compared with group IR, group DEX and group DY showed obviously higher moting function scores,lighter pathological injury,enhanced expressioin of Bax and caspase3, reduced expression of Bcl-2, decreased apoptosis rate of cells(P<0.05). Compared with group DY,group DEX got better outcomes in above items.Conclusion:Dexmedetomidine had neuroprotective effect in spinal cord ischemia-reperfusion injury through activation of α2-adrenoreceptors, phosphorylation of FAK,up-regulation of Bcl-2,down-regulation of Bax and caspase3.
