Objective:To investigate the impact of family history of type 2 diabetes mellitus (T2DM) on pancreatic β-cell function and insulin sensitivity in metabolically healthy overweight/obese individuals.Methods:435 first-degree relatives of patients with T2DM and 116 spouses without family history of T2DM or impaired glucose tolerance were selected. The individuals were divided into six groups:metabolically healthy normal weight of first-degree relatives (FMHNW), metabolically healthy overweight/obese of first-degree relatives (FMHO), at-risk overweight/obese of first-degree relatives (FAO), metabolically healthy normal weight of spouses (the control group, SMHNW), metabolically healthy overweight/obese of spouses (SMHO), at-risk overweight/obese of spouses (SAO) according to body mass index (BMI) and metabolic status. We used homeostasis model assessment (HOMA-IR) and glucose disposition index (DI1 and DI2) to evaluate insulin sensitivity and pancreatic β-cell function, respectively.Results:(1) In the FMHO group, HOMA-IR was higher compared to the FMHNW group (P<0.05), and lower compared to the FAO group (P< 0.05). Simultaneously, DI1 was lower compared to the FMHNW group (P < 0.05) and higher compared to the FAO group. DI2 was higher significantly when compared to the FAO group, but no statistical significance compared to the FMHNW group (P<0.01).(2) When comparing with the SMHO and SMHNW groups, HOMA-IR in the FMHO group was higher (P< 0.05). However, no statistical significance was found in DI2, and DI1 was only found significant when comparing with the SMHNW group. Comparing the FMHNW and SMHNW group, there were no statistical significance in HOMA-IR, DI1 or DI2.Conclusions:Family history of T2DM may affect insulin sensitivity of MHO individuals from the first-degree relatives.
