Objective:To discuss the discrepancy and effects of the expression of ca2+-sensing receptor in pulmonary artery of chronic hypoxia and monocrotaline establishing pulmonary hypertension model in SD rats.Methods:The male SD rats were randomly divided into control group(CON), chronic hypoxia(CH) and monocrotaline(MCT) establishing pulmonary hypertension group.CON group were normally bred, the Rats of CH group were placed in the glass box continued with 10% oxygen made by ourselves, rearing 21 days.MCT group,a dose of 60mg/Kg(BW) MCT were given to SD rats by single intraperitoneal injection, rats were feed in SPF grade for 21 days.We measured:(1) Mean pulmonary arterial pressure (mPAP) were monitored by right heart catheterization.(2) Right Ventricular Hypertrophy Index(RVHI).(3)The pathological changes of lung tissue were stained by Hematoxylin and Eosin(HE).(4) Expression of Ca2+-sensing receptor (CaSR) mRNA and protein detected by QPCR and Western blot.Results:Comparison of the CON group, the mPAP and RVHI increase in CH and MCT group with the extending of intervening time, CH 21d and MCT21d were the highest group(P<0.05). Rats in MCT group were significantly higher than CH group(P<0.05). Expression of CaSR mRNA in experimental 21d group(CH and MCT) were significantly higher than CON group, P<0.05),and the MCT 21d group were higher than CH 21d group(P< 0.05). Expression of CaSR protein increase in experimental group with the extending of intervening time, MCT 21d was the highest group.Conclusion:Pulmonary artery pressure in MCT group is higher than CH in pulmonary hypertension model in rats as the same time. Expression of CaSR mRNA and protein in experimental group(CH and MCT) were higher than control group, CaSR participated in the occurrence and development of PH.
