Chapter 1:This chapter briefly introduced the structure, properties, preparation, purification and biological activities of flavonoids and flavonoids in buckwheat. The preparation and applications of β-cyclodextrin ( β-CD) and the preparation method of inclusion complex were reviewed in detail.Chapter 2:The inclusion interactions of β-CD for rutin have been evaluated based on phase-solubility method by using β-CD as a drug carrier and rutin as a model drug. Before and after the inclusion, the solubility of rutin changes obviously. When the concentration of β-CD is 9 × 10-5mol/L, the solubility of rutin was increased to 5.5 × 10-4mol/L from 2.0 × 10-4 mol/L, increasing 2.7 times. The existence of β-CD will increases the stability of inclusion compounds.Chapter 3:The solubility of rutin was increased by forming complex with cyclodextrin. Fluorescence spectroscopy method was used to evaluate the influence of pH value, common metal ions and common amino acids on the complexation of rutin with cyclodextrin. When the pH value was 4.5 to 5, the properties of inclusion compounds was best, the inclusion constant K was 420M-1 ±10. There is interaction with tryptophan、histidine and phenylalanine when complex was formed, in comparison with other amino acids. The fluorescence of rutin increased with the addition of Al3+. Because rutin coordinated with Al3+ to form chelate, and the ratio is 3:2.Chapter 4:Rutin and β-cyclodextrin inclusion complex were prepared with encapsulation. Then morden methods (Fourier transform infrared spectrum (FT-IR) analyze, Nuclear Magnetic Resonance) and molecular dynamics simulation method were adopted to confirm Rutin, β-cyclodextrin and the inclusion compound. Nuclear Magnetic Resonance indicated that the benzene ring of rutin (B ring) was inserted into the small end of the β-CD to form complex. Moreover, this was similar to that observed from the molecular dynamics simulation method.
