Objective:Intraperitoneal injection of oxygen of oxazine acid potassium was used to produce HUA model in rats.To observe the effects of the different serum uric acid levels on expression of A β 1-42,iNOS,APP and BACE1 in hippocampus of rats. To explore the relationship of the different serum uric acid levels between alzheimer’s disease.Methods:Wistar rats were divided into normal control group, low dose group, the dose and high dose group, and adaptive breeding a week later, given different oxygen concentrations of oxazine acid potassium intervention, respectively 0,2,4 weekend in experiments using tail take blood, blood was collected 1 ml, the natural coagulation at room temperature and centrifugal (3000 r/min,15 min), the detection of serum uric acid level; 4 weekend put to death animals in experiments, the part quickly strip the hippocampus brain tissue, placed immediately-80 ℃ refrigerator, partial strip brain tissue to glutaraldehyde fixation -epoxy resin embedding treatment, conventional fixed-paraffin embedding treatment; H&E staining to observe histomorphology changes rats, immunohistochemical method for determining rat hippocampus Amyloid--Protein(1-42) (Aβ 1-42) and type induced the expression of nitric oxide synthase (iNOS); Western blot detection hippocampus amyloid peptide precursor protein (APP) and Beta-site amyloid precursor protein cleaving enzyme-1(B ACE 1) expression.Results:Results show that high uric acid hematic disease rat model building success, low serum uric acid level, middle and high dose group were higher than that of normal control group, the result was statistically si gnificant (P< 0.01). Immunohistochemical results show that the high uric acid hematic disease model rats brain hippocampus A beta 1-42 and obviously up-regulated the expression of iNOS than normal control group, and with the increase of serum uric acid level, A beta 1-42 and the expression of iNOS is on the decline. Western blot analysis result, low, medium and high dose group of the expression of rat hippocampus APP and BACE1 is negatively related to the level of serum uric acid and low dose group of highest expression of rat hippocampus APP.Conclusion:The higher can lead to the occurrence of AD than normal serum uric acid levels, the risk of AD was falling with the increase of serum uric acid level.
