Lotus plumule is the green radicle in the middle of Nymphaeaceae Nelumbo nucifera Gaertn. ripe seeds,which is widely distributed in Hunan, Hebei, Guangdong, Fujian, Jiangxi and other provinces.Lotus plumula processes the functions of lowering blood pressure, lowering blood sugar, anti-inflammatory, anti-cancer, anti-bacterial,anti-anxiety and so on. Main chemical composition of Lotus plumula are alkaloids, as well as flavonoids, polysaccharides and volatile oil compounds. Numerous studies show that the compounds isolated from the traditional Chinese medicine has good efficacy and low toxicity and it is a very important food and medicine isogeny object. In this paper, the isolation, purification and identification ofchemical constituents in lotus plumula, and the purification process of neferine from lotus plumula, were studied for the purpose of exploiting anti-inflammatory drugs.1. The chemical constituents in lotus plumula were isolated, purified and identified. Reverse silica gel column chromatography, ODS column chromatography, polyamide column chromatography, Sephadex LH-20 column chromatography, high performance liquid chromatography (HPLC), recrystallization and other chromatographic separation methods were comprehensively utilized and 8 compounds were isolated from chloroform extracts of ethanol extract from lotus plumula. The structures of them were identified by mass spectrum and other spectrum analysis methods. Five of compounds were alkaloids:Liensinine(1), Isoliensinine(2), Neferine (3), Nuciferine(4) and Higenamine(5). Three of them were flavonoids:Rutin, (6), Hyperoside(7), and Luteolin-β-D-glycosidase (8), and Compound 8 is isolated from Nymphaeaceae plant for the first time.2、The flash extraction process of total alkaloids from lotus plumula was studied first, and then the macroreticular adsorption resin preparation process of neferine was studied. The extraction process of total alkaloids from lotus plumula was optimized by using single factor and orthogonal experiment. The extraction solvent, extraction time, extraction times, the amount of solvent and other aspects of the extraction process was optimized. The optimum extraction process was:17.74 times (w/w) of 60% ethanol was used as the extract solvent, the extraction time was2.76min and the raw materials were extracted three times. Neferine was enriched with macroreticular adsorption resin, and macroreticular adsorption resin D101 type was selected by measuring different kinds of resin adsorption rate and desorption rate. The elution solvent, sample liquid pH value, elution water amount, eluent amount, loading amount, elution flow rate and other influence factors was screened. And the best macroreticular adsorption resin purification process condition is that:The elution solvent was 60% ethanol, pH value of the loaded sample was 9, the elution water amount was 4Bv, the eluent amount was 4 to 5 Bv, the loaded sample amount of the resin (g)/medicinal materials (g)=1/0.4, and the elution flow rate was 2 Bv/h. Then the eluent products we get were spin steaming enriched, dried under reduced pressure, and re-dissolved by methanol solvent. The neferine content in the end products was 70.0% determinated by HPLC.The results show:Neferine alkali content increased from 16.9% to 70.0%, the purity increased by about 3.3 times.
