Nephropathy DN(diabetic) is a common chronic complication caused by diabetes mellitus,which can lead to end-stage renal disease mellitus,and it is one of the most serious complications of diabetes mellitus. The imbalance of ECM synthesis and degradation because of the diabetic nephropathy, resulting in the accumulation of ECM,which is associated with the Metalloproteinases Matrix(MMPs) system, the fibrinolytic system and so on. Schisandra chinensis is dry ripe fruit of Family of Magnolia or Central China Schisandra chinensis, is a traditional Chinese medicine. On the effect of single herb medicine and its extract on diabetic nephropathy, no study has been studied.In this study, we observed the effect of the extract of Schisandra chinensis on the kidney of diabetic rats,and detected the effect of extract of Fructus extract on blood glucose, blood lipid, renal function, oxidative stress and so on. To provide a theoretical basis for the prevention and treatment of DN, the mechanism of the deep mechanism of the matrix metalloproteinase was discussed and to observe the intervention of the extract of Schisandra chinensis on the matrix metalloproteinase system in diabetic rats.Single intraperitoneal injection STZ(55 mg/kg) was used to induce diabetic nephropathy in rats. The diabetic rats were randomly divided into normal control group(Control), model group(DN), Sch group and Benner Pury positive group and to observe the effect of on DN rats after 12 weeks. We detected the blood and urine biochemical parameters and oxidative stress index of each group, and observed the renal pathological changes and PAS accumulation in rats by ECM and HE staining. The expression of FN, Col, TIMP-2 and MMP-2 in kidney was detected by immunohistochemistry; MMP-2 activity was detected by enzyme spectrum method.The results showed that the blood glucose of rats in the DN group was increased, the renal function was significantly changed, and the kidney appeared ECM concentration and other pathological changes. To detect the serum lipid index, the contents of T-CHO and LDL-C in the DN group were significantly increased(P<0.01, P<0.05). Compared with the DN group, Sch group LDL-C significantly decreased(P<0.05), and T-CHO was also decreased. On the oxidative stress index, the activity of GSH-PX and MDA in the DN rat serum were decreased, and the activity of GSH-PX and MDA in kidney tissue was decreased, and the CAT increased, and both was statistically significant(P<0.05), Compared with the DN group, the serum MDA was decreased(P<0.05), CAT, SOD activity was increased(P<0.01), and the MDA was decreased(P<0.05) in the Sch rats. Glomerular lesions of Sch group of DN rats were eased, The proliferation of mesangial cell loss, basement membrane thinning, ECM collagen decreased,the similar effect of bena group. The expression of Col Ⅳ and FN IV was significantly decreased, The TIMP-2 expression of ECM was significantly decreased after Immunohistochemistry experiment. With enzyme spectrum,MMP-2 activity in diabetic rats was significantly decreased(P<0.05). Compared with the diabetic group,the MMP-2 activity in the Sch group was significantly enhanced(P<0.05).According to the research results, the following conclusions are drawn:Effect of extract of Fructus chinensis on renal protection in experimental diabetic rats induced by STZ.The mechanism may be related to reducing blood lipid, improving the level of serum and renal tissue oxidative stress and improving the renal MMP-2 activity, inhibiting the expression of TIMP-2 and improving the degradation of extracellular matrix.Innovation points:In this study, the effect of the extract of Fructus chinensis on the renal function of experimental diabetic rats was observed for the first time, and the possible mechanism of renal protection was discussed from the point of view of ECM degradation enzyme.
