Doxorubicin, as the most common broad-spectrum anti-tumor drug, is widely used in the chemotherapy of acute leukemia, breast cancer, lung cancer, ovarian cancer and so on. But it is often accompanied by serious heart, kidney, liver toxicity. Therefore, finding new preparations of Doxorubicin with low toxicity and high water solubility is still one of the most important issues in the treatment of cancer. In view of the problem of the new preparation of Doxorubicin,this paper designs a kind of Doxorubicin loaded nanoparticles and evaluates its cytotoxicity to breast cancer cells. It provides reference for the exploration of new type of chemotherapy for breast cancer.In this paper, firstly, through bulk polymerization method with D, L-lactide and glycolide as raw material for the synthesis of PEG-PLGA copolymer and application of 1HNMR characterization. PEG-PLGADOX micelles were prepared by dialysis method with adriamycin and PEG-PLGA copolymer, and then investigated the morphology, particle size, drug loading rate, encapsulation efficiency and drug release behavior: The micellar morphology is spherical and the particle size is 135 nm, The drug loading rate was(9.7+0.33)% and Encapsulation rate was(71.63±1.33)%, it showed slow release compared to free drugs in 96 h. The biocompatibility of PEG-PLGA copolymer and cytotoxicity of PEG-PLGA-DOX on 4T1 cell line were investigated by MTT method in vitro. The results showed that PEG-PLGA has negligible cytotoxicity and good biocompatibility; Compared with free doxorubicin, PEGPLGA-DOX significantly reduced the cytotoxicity of doxorubicin; Flow cytometry and confocal laser scanning microscopy were used to detected the cell uptake of DOX micelles. The results of the experiments showed that the uptake of micelles increased with time. The treatment of Tumor-Bearing mice, Histological analyses of tissues, organs and Immunohistochemical and immunofluorescence staining proved PEGPLGA-DOX to reduce the serious side effects of DOX, and significantly improve the anti-tumor effect. PEG-PLGA-DOX is a new type of nanomedcine for the safety and effectiveness of triple negative breast cancer.
