Objective:To study the growth effects of Notch signaling pathway inhibitor DAPT on HER-2 positive breast cancer cells with stem cells properties.Methods:1.Incubate the ALDH1 positive and HER-2 overexpressing breast cancer stem cells in vitro, then divide these cells into two groups respectively, add DMSO as vehicle group and DAPT with different concentration as observed group.2. To identify the stem-cells characteristics of SK-BR3 using Microspheres formation examination.3.To detect the growth inhibition of DAPT on SK-BR3 cells by MTT.4.The flow cytometry was used to assay cell apoptosis.5. Tumor cells migration was measured by Transwell invasion Results:1.SK- BR3 cells can form microspheres in serum-free suspension culture.2.DAPT inhibited the growth of the SK- BR3 cells and the efficacy was dose-dependent.3.DAPT can induce SK- BE3 cell apoptosis and the apoptotic rate is dose-dependent.4.DAPT can reduce the migrated ability of SK- BR3 cell.Conclusion:Notch signaling pathway inhibitor DAPT was of obvious effects on Her-2 positive and rich in stem cells SK-BR3 cell lines in inhibiting growth, promoting apoptosis and reducing invasion. DAPT may be one of the promising therapeutic agent to breast cancer.
