It has been reported that homeoboxa 10 (HOXA10) acts as a transcription factor during embryo implantation, but the regulatory mechanism of its biological function is still unclear. In a yeast two-hybrid screening experiment, we found that the acetyltransferase P300/CBP associated factor (PCAF) interacted with HOXA10, suggesting that PCAF might acetylate HOXA0 during the process in establishing endometrial receptivity.In this study, PCAF was identified as a HOXA10-interacting protein and HOXA10 associated with PCAF through its homo-box domain. PCAF inhibited HOXA10-mediated β3-integrin (ITGB3) transcription through its histone acetyltransferase (HAT) activity (P <0.05). Over-expression or knockdown of PCAF showed PCAF down regulated HOXA10-mediated ITGB3 protein expression by acetylating HOXA10 and significantly diminished HOXA10 mediated embryo adhesiveness in Ishikawa cells (P<0.05).We also found that, there was an aberrant high PCAF expression in the endometrium of women with endometriosis than that in normal controls (P<0.05).In summary, HOXA10 associates with and is acetylated by PCAF in Ishikawa cells and HOXA10-PCAF association impairs embryo implantation by inhibiting ITGB3 protein expression in endometrial epithelial cells. | Keywords/Search Tags: | PCAF, HOXA10, acetylation, Ishikawa cell, EMT, implantation | PDF Full Text Request | Related items |
