Objective:Application of combined MALDI-TOF-MS WCX magnetic bead method to detect the blood protein fingerprint between primary osteoporosis type Ⅰ and normal rat serum, and buid protein fingerprint to screen differentially expressed proteins. Profit how the qiangguyin affect on expressed proteins, thus identify screening and diagnosis of primary osteoporosis type Ⅰ specific molecular markers.Methods:1. Choose 40 female SD rat weighting 220g to 250g,and select randomly 10 rat as control group,while select 10 rat randomly from the remaining rat as experimental group resection of bilateral ovaries. Keep these rats for 10 weeks, then detect bone mineral density value to judge if osteoporosis building is build successfully. Under take 1 ml blood on an empty stomach, then prepare of serum protein WCX magnetic beads, separation and purification of point target, and collect date with MALDI-TOF-MS to invite serum protein fingerprint of homogenization processing and data analysis.Last through the genetic algorithm in combination with support vector machine (SVM) algorithm, screen of different protein peak and select the significant difference of protein peak random combination, of those date the highest program index of protein peak to establish diagnosis model and its protein peak candidate markers for disease.2.4 rat is death for Resection of bilateral ovaries.Remaining 26 SD rats were divided into two groups, of one group giving qiangguyin as experimental group while another group giving normal saline as control group. Respectively six weeks later and two months later, take on an empty stomach blood, and use ELISA method to detect the OrphaninFQ and SelenoproteinW value in osteoporosis model in rat serum matrix. Analysis the date with statistics software spass19.0 and write it in the form of the mean and standard deviation.Results:In the first experiment, four protein peak have significant differences, about an index of the highest Orphanin FQ solitary brown peptide and SelenoproteinW, preliminary diagnosis and screening of osteoporosis as markers.In the second experiment, before breeding Orphanin FQ solitary brown peptide and selenoprotein W had no significant difference, P> 0.05. Six weeks later,experimental OrphaninFQ solitary fei peptide value is lower than the control group, no significant difference, P> 0.05. While experimental group SelenoproteinW value is higher than the control group, then statistical software analysis, being practical significance,P<0.05.Two month later, experimental group OrphaninFQ solitary fei peptide value is lower than the control group, while experimental group SelenoproteinW value is higher than the control group, also exsiting obvious gap, P<0.05, being practical significance.Conclusion:Orphanin FQ and SelenoproteinW may be a marker for diagnosing and screening primary osteoporosis type Ⅰ.Orphanin FQ with Selenoprotein W and qiangguyin probably have some relation in triggering primary osteoporosis type Ⅰ.
