Objective:To discuss whether pretreated with Panax quinquefolium saponins (PQS) can relieve the injury of Ischemia-reperfusion in HK-2cells and its possible mechanism.Methods:HK-2cells were treated with various concentrations (40μg/mL,160μg/mL 640μg/mL) of PQS for 4h respectively. The model of injury of HK-2cells in vitro were induced by hypoxic reperfusion injury for 4h. The expression of Superoxide Dismutase was measured by enzyme-linked immunosorbent assay. The cell mitochondrial membrane potential (△Ψm) was detected by using JC-1 staining method. Flow cytometry were used to observe the apoptosis. Then Western blotting were used to detect Bcl-2 and Bax expression in HK-2cells treated with PQS.Results:PQS used before Ischemia-reperfusion can increased the activitiy of SOD in HK-2cells and has evidence antioxidation (P<0.01). Comparing to the control group, The mitochondrial membrane potential of PQS groups was significantly higher (P<0.01). The apoptosis rate of HK-2cells was significantly decreased after 4h of PQS tratment compared with the control(P<0.01). The expression of Bax was decreased whereas the expression of Bcl-2 was increased by Western blotting analysis after treated HK-2cells with various concentrations(40μg/mL,160μg/mL,640μg/mL) of PQS for 4h, and they all acted in dose-dependent manner.Conclusions:②PQS has the protective effect on myocardial ischemic reperfusion injury in HK-2cells.②The down-regulation of Bax and the up-regulation of Bcl-2 may related to PQS-induced anti-apoptosis of HK-2cells. |
