Objective:The issue in vitro experiments, is to observing Sinomenine effect on ox-LDL stimμLated THP-1 derived foam cell cholesterol metabolism, NF-KB activition and release of inflammatory cytokines IL-1β, IL-8, MCP 1-secretion,through which to further explore Sinomenine prevention of atherosclerosis possible mechanism. Grouped given different drug-containing medium, with the control group to complete medium,for 12 hours,24 hours and 48 hours, the supernatant was collected and cells were within the enzyme assay to detect cell changes in cholesterol; detected by ELISAIL-1β, TNF-α and MCP-1 and the amount of NF-KB activity.Research design and methods:In THP-1 experimental objects, Group seven groups: control group (complete medium), different concentrations Sinomenine group (1ug/ ml,20ug/ml,40ug/ml,80ug/ml),5ug/ml simvastatin group and 100ug/ml to plug betamethasone group. To establish AS foam cells model by ox-LDL stimμLating THP-lcell for 48 hours.ResμLts:1. Sinomenine has certain inhibition in foam cell growth, when less than 100ug/ml obvious. While significant inhibition, or even death,when more than 100ug/ml.2. Compared with the control group, cholesterol drug group can significantly reduce inflammatory cytokines IL-1β, IL-8, MCP-1 levels, with difference was statistically significant (P<0.01). And the role has the more obvious trend with time and concentration increasing.3. In addition to Sinomenine 1ug/ml group, each group has lower NF-KB content than the controled group. Difference is significantly at 24-hour time point and 48-hour time point (P<0.01 or P<0.05), In contrast to 12-hour time point (P> 0.05).4. Tthe sinomenine group’s IL-1β、IL-8 content decreased more those of simvastatin group in 24-hour time point, with a significant difference (P<0.05). Compared with simvastatin Sinomenine group’s MCP-1 content has more tendency to decrease, but no significant difference (P> 0.05).5. Compared with the dexamethasone group, Sinomenine group of inflammatory cytokines IL-1β、IL-8、MCP-1 and NF-KB content reduced less, with significant difference (P<0.01 or P<0.05),;while cholesterol content has no significant difference between each other(P> 0.05).Conclusions:1. Sinomenine may inhibite cell growth was with increasing concentration of the more obvious.2. Sinomenine inhibits foam cell inflammation by inhibiting the activity of NF-KB, the release of inflammatory cytokines IL-1β, IL-8 and MCP-1, reducing cholesterol metabolism, and thus to improve atherosclerosis. Its anti-inflammatory effect is weaker than dexamethasone but stronger than simvastatin.
