| Abstracts:objective:Penthori Herba is also called ganhuangcao,shuizelan and shuiyangliu.Penthori Herba is the dry above-ground parts of Penthorum chinense Pursh. (Fam.Saxifragaceae).It has the effect of clearing heat and detoxicating,removing jaundice and resolving dampness,activating blood and dissipating blood,clearing damp and promoting diuresis. Penthori Herba is more effective in the treatment for each hepatitis. Penthori Herba distribution around above 1000m coming foothills virgin forests. Its authentic region in GuLin Luzhou. Because there is no normative quality standards of Penthori Herba, which influence its medicine quality and curative effect.This topic will research pharmacognosy of Penthorum chinense Pursh,and identificating its active ingredients by TLC, and compare the active ingredients content in different medicinal parts, and provide the basis for pharmacognosy research of Penthorum chinense Pursh. Methods:This study used the original plant identification, trait identification, microscopic identification and chemical identification method to make a Pharmacognosy Identification. Thin Layer Chromatography (TLC) was used for qualitative identification of quercetin and gallic acid. Ultraviolet spectrophotometry was used to determine total flavonoids content in different medicinal parts,and high performance liquid chromatography (HPLC) was used to determine the content of quercetin and gallic acid in different medicinal parts of Penthorum chinense Pursh. Results:There are obvious characteristics in original plant morphology, microscopic characteristics, trait characteristics and chemical characteristics. According to the appendix method in the pharmacopoeia of the People’s Republic of 2010 version, determining the extract content, water content, ash content, heavy metal and organic pesticides residues content. Experimental data shows:the extract alcohol-soluble using the cold dip method to determine the average content of 27.06%,and using the hot dip method to determine the average content of 23.84%,the average moisture content is 8.45%, the total ash content is 7.42%, the acid insoluble ash content is 0.73%, the tresidue on ignition is 9.16%,heavy metal check is qualified, the organic pesticides residues not be detected;quercetin and gallic acid could be detected by thin layer chromatography (TLC);the total flavonoids content were determined by ultraviolet spectrophotometry, total flavonoids was good linearity in 0.0058-0.0928mg·ml-1, the scope relationship was 0.9996, the average recovery was 98.13%, and RSD was 0.81%, the test shows total flavonoids content is 2.17% in fruit,1.30% in stem,3.59% in leaves,2.65% in above-ground part; the quercetin content were determined by high performance liquid chromatography (HPLC), quercetin was good linearity in 0.0088~0.088mg·ml-1, the scope relationship was 0.9999, the average recovery was 101.94%, and RSD was 0.72%, the test shows quercetin content is 0.0671% in fruit,0.0075% in stem, 0.0709% in leaves,0.0572% in above-ground part;the gallic acid content were determined by high performance liquid chromatography (HPLC), gallic acid was good linearity in 0.0042~0.1060mg·ml-1, the scope relationship was 0.9999, the average recovery was 98.02%, and RSD was 1.18%, the test shows gallic acid content is 0.4467% in fruit,0.1939% in stem,0.4816% in leaves, 0.3898% in above-ground part; Conclusion.This experiment establishes the pharmacognostical identifications for Penthorum chinense Pursh. The TLC shows Penthori Herba contains quercetin and gallic acid composition; This experiment establishes a way for using ultraviolet spectrophotometry to determinate the total flavonoids content in different medicinal parts, this way is simple, rapid, sensitive, high precision, high accuracy and with good repeatability and recovery; this experiment establishes a method for using HPLC to determinate the content of quercetin and gallic acid, the method i,s. simple, high precision, high accuracy and with good repeatability and recovery. Content of quercetin and gallic acid in the leaves of Penthorum chinense Pursh is the highest. |
