Objective:To study the protective effect of bezoar detoxification on rat model of acute lung injury induced by lipopolysaccharide inHIF-1a,superoxide-dismutase(SOD),and explore its possible mechanism of protection.Methods:72 male SD rats, randomly divided into normal saline control group (group A), lung injury model group (group B), bezoar pretreatment group (group C),24 in each group; Theneachgroupwererandomlydividedinto3groups,namelyA1,A2,A3,B1,B2,B3,C1,C2,C3,a nd a total of 9groups.The tail vein of rats in the blank control group (group A) were injected 5ml physiological saline erery kilogram body weight; The tail vein of rats in the model group (group B) and in vitro cultivated calculus bovis intervention group(groupC), were injected 5ml lipopol ysaccharide erery kilogram body weight; The Bezoar pretreatment group began gastric perfusion in vitro cultivated calculus bovis 0.lg/kg 14 days before,then take the injection of lipopolysaccharideafter.The blank control group (group A) and model group (group B) were injected with saline of the same dosage. The rats in group A1,B1,C1 were killed after 6 hours; The rats in group A2、B2、C2were killed after 12 hours; The rats in group A3,B3,C3 were killed after 24 hours;Last mesure and calculate the lung wet/dry of all groups.Immunohistochemical method was used to mesure changes in the expression of HIF-la in each group, in order to observe the pathological changes of lung tissue of each group, and compared the results with.groups.Results:(1) The expression change of HIF-1a:The HIF-la expression in groupAl,A2,A3 had no obvious changes;The HIF-la expression in groupB1,B2,B3 obviously increased,and the expression of B2 is the most obvious. The HIF-la expression in group C1,C2,C3were also expressed,and the expression of C2 is the most obvious more than A2,but less than B2;(2) The W/D value changes in rats:The ratio of each time point in group B was higher than groupA,the difference was statistically significant(P<0.05).the ratio of group B and C was lower compared with each other.but still higher than that of groupA,the difference was statistically significant(P<0.05),but higher than the group A.The difference was statistically significant(P< 0.05).(3)The pathological changes in lung tissue of rats:The groupAhad no pathological changes; groupB inflammatory changes obviously,and the B2changes were most obvious; Under the optical microscope, pulmonary congestion, edema, and inflammatory cell infiltration can be observed. The inflammatory changes lessened of group C, and that in group C2 than in group Cl,group C2 and group C3 are basically the same.(4)The expression changes of SOD:The ratio of group B was significantly lower than that of group A,the difference was statistically significant(P<0.05),the ratio of group C and group B are high,but still lower than that in group A,the difference was statistically significant(P<0.05)。Conclusion:Bezoar after pretreatment makes the damage degree of pathological damage of rat lung attenuate acute lung; The value of W/D decreased obviously; The expression of SOD activity increased, The expression of HIF-la was decreased;The interraction between Bezoar and synergistic effects of SOD, avoid the excessive production of oxygen free radicals, improve the body’s ability to scavenge free radicals, improve the ability of hypoxiatolerance, reduce lipid oxidative damage effects on tissue cells;inhibits the HIF-la expression, reduce in the pathogenesis of ALI HIF-la as the amplifying role in the inflammation cascade reaction; ease the inflammatory cell exudation, which offer protective effects on the acute lung injury to some degree.
