Objective: Simulating the processing progress of Chinese materia medica such as our subjects Lotus leaf, Pollen typhae and Rhizoma nelumbinis utilizing the Thermogravimetric(TG) technology, and quantitative processing technology of the traditiona temperature range by thermogravimetric(TG), derivative thermogravimetric(DTG). Comprehensive analysis the changes between "internal medicinal ingredient" and "extrinsic Characters",by instrument analysis technology like HPLC. Initial clarify Lotus leaf, Pollen typhae and Rhizoma nelumbinis fried charcoal processing technology and mechanism, and explore the common law of carbon drug.Methods:① Under the simulated air atmosphere, using thermogravimetric- derivative thermogravimetric(TG- DTG) technology to simulate the TCM firing process.The powder of Lotus leaf, the alcohol extract of Lotus leaf, the total alkaloid extract of Lotus leaf, the total flavonoids extract of Lotus leaf, quercetin reference substance, isoquercitrin reference,quercetin glycosides reference substance, Hyperoside reference substance pyrolysis Characteristics were studied to examine the optimum processing temperature of Lotus leaf carbon..② The powder of Pollen typhae, the alcohol extract of Pollen typhae, the total flavonoids extract of Pollen typhae, the tannin extract of Pollen typhae,quercetin reference substance, typhaneoside reference substance, isorhamnetin-3-O-neohesperidin glucoside reference substance pyrolysis Characteristics were studied to examine the optimum processing temperature of Pollen typhae carbon.③The powder of Rhizoma nelumbinis, the water extract of Rhizoma nelumbinis, the tannin extract of Rhizoma nelumbinis, gallate reference substance pyrolysis Characteristics were studied to examine the optimum processing temperature of Rhizoma nelumbinis carbon.④ Quercetin content in lotus leaf carbon samples under different concocted process were given by High performance liquid chromatography, and with the appearance of the samples to explore the optimum concocted processing of lotus leaf carbon.Results:①quercetin and other flavonoids is the main medicinal ingredient for their relative content increase during lotus leaf carbon process; optimum ceiling temperature rang is320 ° C, the optimum temperature is 280 ° C.②Tannin and flavonoids as the main medicinal ingredient of Pollen typhae charcoal for it’s content increase during the Pollen typhae carbon fried; Pollen typhae main active ingredient studied by pyrolysis characteristics, then make sure 250 ° C was the ceiling temperature of Rhizoma nelumbinis carbon, the optimum processing temperature was 200 °C.③Tannin as the main medicinal ingredient of Rhizoma nelumbinis charcoal for it’s content increase during the Rhizoma nelumbinis carbon fried; Rhizoma nelumbinis main active ingredient studied by pyrolysis characteristics, then make sure 360 ° C is the ceiling temperature range of Rhizoma nelumbinis carbon, the optimum processing temperature was 300 ° C.④ Determination of quercetin, the regression equation y = 36.317x-46.344(R =0.9994), quercetin reference range in 1.274-40.768 ug / m L, good linearity. Good precision instruments used in the experiments, RSD is 1.815; good experimental samplesstability within 12 h,RSD was 3.10%; test methods and good reproducibility, RSD was 4.03%experimental recovery was 95.17-104.72%, RSD was 4.86%. Experimentally measured LC2 quercetin content was the highest one during LC1-LC12 samples, LC4 second, and comprehensive LC2, LC4 samples morphology characteristic, conclusioned, optimal processing technology of carbon lotus was 10 g lotus leaf, 200 o C feeding, control pot temperature of 280 ± 10 o C, stir 8min.⑤ Summary the thermal stability law of the main active ingredient in Lotus leaf,Pollen typhae and Rhizoma nelumbinis: Alkaloid <flavonoid glycoside <hydrolysable tannin <flavonoids <condensed tanninsConclusion:In this study, thermal analysis techniques first introduced to research Lotus leaf, Pollen typhae and Rhizoma nelumbinis charcoal fried processing, principles and the common law of them.And make the research more objective, specific and digitization. The model related outcomes obtained is applied to the Lotus leaf carbon processing research,explore practical of thermal analysis model, proven the feasibility ofthermal analysis technology used in Lotus leaf, Pollen typhae and Rhizoma nelumbinis concocted process, even entire field of the heating processing. The experiment lay the foundation of thermal analysis techniques deeper development in the field of traditional Chinese medicine professing.
