Objective:The present study was undertaken to observe the expression of IL-17, IL- 10 and IFN-γ in human serum and compare the population of circulating CD4+T cells(CD4+CD25high, CD4+CD127lowCD25+ and Foxp3 +CD4+)in women with healthy pregnancy and preeclampsia. The results to be obtained will provide new clue for study in prevention and treatment of preeclampsia.Methods:Peripheral blood samples were obtained from third-trimester, healthy, pregnant patients(n =38) and preeclamptic patients(n=32). The characteristics of preeclamptic participants were summarized according to the standards of Obstetrics and Gynaecology, the 7th edition written by Lejie in 4th hospital of Shijiazhuang. And the control group of healthy, pregnant patients was randomly chosen in the same hospital. Exclusion criteria for all pregnant participants were diabetes mellitus, hereditary diseases, and/or infectious diseases diagnosed in the course of pregnancy. There was no statistic significance in the age and weight index before pregnancy between the two groups. The peripheral blood lymphocytes were isolated and CD4+CD25high,CD4+CD127lowCD25+ and Foxp3 +CD4+ Treg cells were counting by technology of flow cytometry. Serum samples of subjects were extracted from blood samples and then stored at-80 until analyzed. Levels of IL-17, IL- 10 and IFN-γ in serum were measured by standard sandwich ELISA kits(R&D Systems) according to manufacturer’s instructions. S t a t i s t i c a l a n a l y s i s w a s p e r f o r m e d u s i n g S P S S v e r s i o n 1 2. 0.Results:1 In the current study, the percentage of CD4+CD25high,CD4+CD127lowCD25+ and Foxp3 +CD4+ Treg cells was 1.02±0.05%, 11.94±1.63% and4.88±0.82% in the CD4+T cells of the circulating blood of the healthy pregnancy. The percentage of CD4+CD25high,CD4+CD127lowCD25+ and Foxp3 +CD4+ Treg cells was 0.54±0.13%、5.74±0.85% and 2.67±0.41%in the CD4+T cells of the circulating blood of the preeclampsia. The proportion of CD4+CD25high, CD4+CD127lowCD25+ and Foxp3 +CD4+ Treg cells was signi?cantly decreased in preeclampsia compared with healthy pregnancy(P<0.05).2 In the current study, the expression of IL-17、IL- 10 and IFN-γ was 38.95±4.50, 21.65±2.33 and 81.89±14.28 pg/ml in the serum of normal pregnancy, while it changed to 569.63±26.21, 13.24±3.59 and 116.58±11.21 pg/ml in the serum of preeclampsia group. The expression of IL-17 and IFN-γ was significantly elevated(P<0.05) and IL- 10 was reduced(P<0.05) in p r e e c l a m p s i a g r o u p c o m p a r e d w i t h h e a l t h y p r e g n a n c y g r o u p.Conclusion:1 The increased expression of IFN-γ and IL-17 may plays a key role in process of a successful pregnancy.2 The effect of cell immunity inhibition and humoral immunity elevation conducted by IL-10 may plays a key role in process of a successful pregnancy.3 The expression of Treg cell was significantly increased in healthy pregnancy, which played an important role in maintenance of a normal pregnancy and f o r m a t i o n o f i m m u n o l o g i c t o l e r a n c e b e t w e e n t h e m a t e r n a l a n d fetal medium.
