Objective:Now tuberculosis morbidity and mortality is increasing gradually, the species of resist antanti-tubercolsis drugs increased year by year,which led to the difficulty of treatment increased and had brought the huge economic burden to the society,.Now,the researchers at home and abroad are committed to looking for new ways to study the new anti-tuberculosis drugs and vaccines.Which purposes are to decrease the morbidity、 increase cure rate and reduce the number of TB deaths. Myeloid differentiation factor 88 (Myeloid differentiation factor 88, Myd88) is an important protein transduction in toll-like receptors signaling pathways, and play an essential role in the signal path, can be directly or indirectly active the innate immunity and adaptive immunity.During the process of anti-tuberculosis immune respose,the cytokines also play an important role. Now research suggests that Myd88 has played important role in resistance Mycobacterium tuberculosis (Mycobacterium tuberculosis, MTB) infection. In this paper,We are to exprore Myd88 and IL-12、IFN-γ in the expression of different crowd tuberculosis,and whether Myd88 could regulate the seretion of IFN-γ、IL-12,that provide the evidence for searching for new ways to treat tuberculosis.Methods:1.This experiment is divided into three groups:the experimental group contains 40 cases tuberculosis patients;the control group contain 38 cases of stable tuberculosis patients;the normal group contain 45 cases healthy people.2.Real time PCR method to detect the expression of three groups of Myd88mRNA.3. EL AS A method were used to detect the three groups of IL-12 and IFN-y.4.We decided to use ST2825 interfere Myd88, ELASA method to detect three groups of IL-12 and IFN-γ level changesResults:1.Experimental Myd88 gene expression quantity is significantly higher than the control group and normal group, said the gene expression level was obvious rise, with statistical significance (P< 0.05), the control group and blank group Myd88mRNA expression quantity, though there are differences, but without statistical significance.2.The level of IL-12 and IFN-γ is respectively 2135.3210±45.088 and 2135.3210 ±45.088 in experimental group;The level of IL-12 and IFN-γ is respectively 953.0217±13.3146 and 769.0133±17.0042 in control group;level of IL-12 and IFN-gamma is respectively 748.0338±19.4030 and 599.0024±15.7015 in normal group.These data in three groups have obvious differences, which has statistically significant(P< 0.05).3.Compared to before ST2825, experimental group of IL-12 and IFN-gamma secretion levels are significantly reduced after ST2825,these data have obvious differences, which has statistically significant (P< 0.05), while the control group before and after adding ST2825 IL-12 and IFN-y change in the level of gamma, but no difference, the data is not statistically significant.Conclusions:1. Myd88 were expressed in the experimental group、control group and normal group,it is says that MTB can stimulate the activation of Myd88 and to induce the rise. Which shows that Myd88 participate in anti-tuberculosis immune response.2. The three group has differernt level of IL-12 and IFN-γ,it is that cytokines play a protective role in the process of anti-tuberculosis immune.3. We The inhibition of the success of the experiment to be ST2825 Myd88, Compared to before ST2825, the level of IL-12、IFN-γ in experiment group and contrast group decreased, The above shows IL-12 and IFN-γ has relationship with Myd88.
