| Background:Malignant tumor is harm to people’s life and health is one of the most common and the most serious disease. Chemotherapy and radiotherapy has been occupies the main status in the treatment of malignant tumor. However, in the process of treatment, chemotherapy and radiotherapy not only kill cancer cells, cells of the body’s other normal tissue also has a lot of damage, and lead to different degrees of side effects. Such as bone marrow suppression, especially on bone marrow hematopoietic stem cell damage is particularly serious, resulting in a decline in number of bone marrow-derived peripheral whole blood cells. Especially in the health care crisis is most severe in white blood cells and platelets, and then there will be a fatal infection and hemorrhage. Because of those, the process of chemotherapy and prognosis of survival in patients will have a negative impact.At present, for the bone marrow cells injury by tumor chemotherapy, mainly rely on drugs increased leukocyte and platelet transfusion and the related factors to improve patients treated. However, these treatments have side effects for some patients even invalid, and expensive. We need to find cheaper than existing treatments, fewer side effects of treatment methods to improve or auxiliary improve these adverse reaction after chemotherapy.In recent years, as the more popular a kind of safe and effective method of physical therapy, low intensity pulsed ultrasound (LIPUS) appears in various kinds of animals and clinical trials. Many reports findings LIPUS can promote mesenchymal stem cells, fibers, etc a variety of cell proliferation. Our preliminary research has been preliminary confirmed LIPUS irradiation can promote normal bone marrow nucleated cell proliferation, and the source of the white blood cells and bone marrow nucleated cells proliferation closely linked. Therefore, this experiment using cytarabine processing New Zealand rabbit, and LIPUS treatment animal model is adopted to observe bone marrow-derived blood cells changes in peripheral blood. LIPUS whether can be used to improve bone marrow cells injury by chemotherapy in the further research.Objective:1. To observe the influence of low intensity pulsed ultrasound (LIPUS) with irradiation for 7 days on bone marrow cells injury by Ara-C in New Zealand rabbit. To observe the rabbit’s general situation and the change of bone marrow-derived blood cells routine.2. To detect part platelet activation function changes after LIPUS irradiation for 7 days on bone marrow cells injury by Ara-C in rabbit.Methods:1. Injected Ara-C into rabbits via the ear marginal vein, dose of 50 mg/kg, for three days. During the modeling and after the modeling, take the rabbit ears arterial blood for blood routine examination.2. The rabbits in model group were randomized divided into LIPUS group (n=10) and control group (n=10). LIPUS irradiation group was carried out irradiation for 7 days on the model of rabbit. The irradiation area of rabbit was the femoral epiphysis end. Waxing the skin before irradiation. Irradiation parameters of 0.2 W/cm2, frequency 0.6 MHz, time to 20 min. Control group using false irradiation, the operation is the same with LIPUS irradiation group, but not open the power supply. Regular blood tests before and after irradiation respectively, until 21 days after irradiation. Counted the cell number of WBC, RBC, PLT, NE, LY, MO. Tested the content of TXB2 and GMP-140 in the serum.Results:1. After Ara-C injection, the cell number of white blood cells and platelets of rabbits in model group were decreasing. After 3 days white blood cells decline has significant difference compared with before the Ara-C injection(P<0.05). Platelets decline has significant difference compared with before the Ara-C injection(P<0.05). Red blood cells decline has no significant difference compared with before the Ara-C injection(P>0.05).2. LIPUS irradiation treatment after 3 days, there was no statistically significant difference LIPUS irradiation group and control group (P>0.05), irradiation after 5,7,11,14,17,21 days, a marked increase in the cell number of white blood cells of LIPUS irradiation group, compared with the control group were statistically significant difference (P< 0.05). Different of red blood cells between LIPUS group and control group had no statistical significance(P>0.05). LIPUS irradiation treatment after 5 days, the platelet count of LIPUS group and control group both in a marked increase arrived a peak. The platelet count in the two groups on day 7 all decreased. The platelet count of 21 days in LIPUS group increase again.3. ELISA test of serum TXB2 and GMP-140 levels of rabbits in LIPUS group and control group before and after Ara-C injection,3,7 days after LIPUS irradiation. Differences had no statistical significance between LIPUS group and control group.Conclusion:1. LIPUS could increase the cell number of NE to prompt the cell number of WBC, and this might be a reason what it could stimulate the proliferation of bone marrow nucleated cells.2. Low intensity pulsed ultrasound had no effect on the number of platelets and red blood cells, the serum TXB2 and the serum GMP-140. |
