Objective:TO investigate the effects of heme oxygenase-1(HO-1) on spiral ganglion cells neurons(SGNs) in cisplatin-induced injury.Methods:Forty adult guinea pigs were divided randomly into 4 groups (n=8 in each group). Group A:To receive intraperitoneal injection of 0.9%saline (12ml/kg,bid) for 3 days. Group B:To receive intraperitoneal injection of cisplatin (12ml/kg,bid) for 3 days. Group C:To receive intraperitoneal injection of ferulic acid (150mg/kg,bid) and cisplatin (12ml/kg,bid) for 3 days. Group D:To receive intraperitoneal injection of ferulic acid (150mg/kg,bid), zinc-protoporphyrin-IX (15umol/kg,bid) and cisplatin (12ml/kg,bid) for 3 days.The expressing pattern of HO-1 in damaged cochlea was studied by immunochemistry and Western Blot technique.Results:, in different groups, HO-1 expression is different in the cochlear spiral ganglion. Group A, The expression of HO-1 is faint in spiral ganglion tissue and its protein expression was 0.48 ± 0.07.Group B,Its protein expression was 0.81 ± 0.07.Group C, Its protein expression was 0.9 ± 0.05.Group D,Its protein expression was 0.61 ± 0.07.The differences between 4 groups were statistically significant (p<0.0001).Conclusion:The expression of HO-1 is maximum in Group C, followed in group B,at least in group C.It shows that HO-1 may be involved in the repair process of spiral neurons damaged by cisplatin.
