Objective:To expolre the NDRG2/Zc3h12d mediated mechanism of the regulation of neutrophil nuclear transcription factor-KB.Method:16HBE cells were stimulated by lipopolysaccharide(LPS), and treated with overexpressed NDRG2 and Zc3h12d siRNA. Cells were divided into 4 groups:A:cells treated with ransfection of overexpressed NDRG2 plasmid;B:cells treated with ransfection of overexpressed NDRG2 plasmid and Zc3h12d siRNA;C:cells treated with ransfection of Zc3h12d siRNA;D:cells treated with ransfection of empty plasmid;E:a control group(without any treat to the cells).Reverse transcription-polymerase chain reaction(RT-PCR) was used to measure the transcription level of mucin(MUC)5AC.The MUC5AC and the inflammatory factors secreted in supernatant were evalued by enzyme-linked immunosorbent assay(ELISA). The expression of NF-κB protein and the protein levels of Zc3hl2d were measured by Western blot.Result:with the stimulation of LPS,the expression of MUC5AC mRNA and MUC5AC protein were higher than those in the control group(p<0.05).The expression of MUC5AC mRNA and MUC5AC protein in overexpressed NDRG2 group were significantly less than those in empty tranfection group(p<0.05).But the expression of MUC5AC mRNA and MUC5AC protein in overexpressed NDRG2 with Zc3h12d siRNA group were high compared with the overexpressed NDRG2 group(p<0.05).Conclusion:NDRG2 is related to Zc3h12d.NDRG2 can regulate the function of NF-κB by Zc3h12d.
