Objective To investigate the effect of curcumin on migration of human umbilical vein endothelial cells induced by ox-LDL.Methods The HUVECs were cultured in vitro for 24h, then the ox-LDL was incubated with the HUVECs to make the redox injure model. Cells were treated with ox-LDL in different concentrations (0,10,20,40, 80,160mg/L) for 24h, the apoptosis rate was determined by flow cytometry. The cells were divided into four groups:①normal control group:cells were treated in normal condition;②urcumin treatment group:(curcumin, 25μmol/L,24h); ③ox-LDL treatment group:(ox-LDL,20mg/L,24h); ④curcumin pretreatment+ox-LDL treatment group:curcumin (25μmol/L, 2h) pretreatment before ox-LDL treatment (20mg/L,24h). The concentration of ROS was measured by laser confocal microscopy; the level of NO was dectected by method of colorimetry; the protein of eNOS, p-eNOS(Ser1177), Akt, p-Akt(Ser473) and ICAM-1 were measured by Western blot; the ability of endothelial cells migration was detected by transwell method.Results After treated with ox-LDL at high concentrations(≥40mg/L), the cell viability decreased remarkably (P<0.05). Compared with control group, the levels of ROS and ICAM-1 in ox-LDL treatment group increased (3.20 ±0.19 vs 1.00 ± 0.06, P<0.01; 2.58 ± 0.28 vs 1.00 ± 0.18, P<0.05), while the phosphorylation of Akt(Ser473) and eNOS(Ser1177), the concentration of NO and number of migration decreased significantly (0.28 ± 0.04 vs 1.00 ± 0.09, P<0.05; 0.37 ± 0.07 vs 1.00 ± 0.05, P<0.001; 0.21 ± 0.057 vs 1.00 ± 0.054, P<0.01; 16.75±2.58 vs 40.69±6.46, P<0.001). Compared with ox-LDL treatment group, the levels of ROS and ICAM-1 in curcumin+ox-LDL group up-regulated obviously (1.47 ± 0.19 vs 3.20 ± 0.19, P<0.001; 1.36 ± 0.28 vs 2.58 ± 0.28, P<0.05),while the phosphorylation of eNOS (Ser1177) and Akt (Ser473) decreased significantly (0.74 ± 0.05 vs 0.28 ± 0.04, P<0.05; 0.88 ± 0.17 vs 0.37 ± 0.07, P<0.01); the concentration of NO and the ability of migration increased significantly(0.79 ± 0.05 vs 0.21 ± 0.06, P<0.05; 27.72±5.36 vs 16.75±2.58,P<0.05).Conclusion Curcumin inhibites the production of ROS induced by ox-LDL, increases the phosphorylation of Akt (Ser473) and eNOS(Ser1177) as well as the level of NO, and promotes the ability of endothelial cell migration.
