Objective:To detect and compare the content of SOD, CRP AND cTnl in the serum, the myocardial no reflow area and myocardial infarct size by setting up AMI reperfusion and Postconditioning models in the purpose to provide more theoretical basis for postconditioning using on clinical test.Method:Thirty rabbits(healthy male rabbit, weighting between 2-3 kg, clean bred, raised in Qingdao Kang da industry Co. Ltd.) were randomly divided into 3 groups:10 in control (Sham) group,10 in surgery group (R/I) and 10 in ischemic postconditioning group (IPOC), The IPOC groups were set up AMI reperfusion and postconditioning models (ligate coronary artery for 1 hour, and then reperfuse for 2 hours). Set up AMI reperfusion models byligating for 1 hour and then dredging the coronary again for 2 hours of R/I group. Take the IPOC group to set up postconditioning models by ligating for 1 hour and then taking dredging for 30s/liagting for 30s as a cycle and do this cycle for 3 times 1 minute before dredging the coronary, and then dredging for 2 hours. For sham group we only do threading without ligation, AMI and reperfusion and observe for the same time as other groups. Coronary artery ligation before 5 min,1 h and after 2 h reperfusion in the carotid blood, using enzyme-linked immunosorbent assay (ELISA) to detect the content and express of serum superoxide dismutase (SOD), high-sensitivity C-reactive protein (CRP) and I(cTnI). To figure out the ischemic region, no reflow area and infarction area by the myocardial pathology; figure out the ligation area, according to the ligation myocardial area/left ventrcular myocardial area and figure out the area of no reflow by no reflow area/ligation myocardial area after the operation.Result:(1) The area of IPOC group of no-reflow and infarct was significantly smaller than the range of R/I, the difference was statistically significant (P<0.05). (2) 5min before ligation in each group of SOD, CRP levels showed no significant difference (P> 0.05); (3) 1,2 hour after ligation and reperfusion in R/I group, IPOC group compared with Sham group, the differences were statistically significant (P <0.05); 1h after ligation IPOC group and R/I group, the difference was not statistically significant (P> 0.05); while at the end of reperfusion 2h IPOC group and R/I group, the difference was statistically significant (P<0.05). (4) 1 hour after ligation IPOC AND R/T group, the ischemia myocardial area and the myocardial infarction area showe no significant difference (P>0.05) while the R/I and IPOC group differences were statistically.Conclusion:(1) Ischemic postconditioning can reduce the damage of the rabbits after AMI reperfusion, it also reduces the area of myocardial infarction, the Degree of myocardial necrosis and then to reduce the occurrence of no-reflow in the end. (2) Ischemic postconditioning may also plays a role on Oxidation and reducing the inflammation after AMI reperfusion.
