Objective: To examine Prion protein(Pr Pc) expression and its clinical significance in Nasopharyngeal carcinoma and analyze the results with the corresponding correlation between clinical parameters. Method: The expression of Pr Pc in Nasopharyngeal carcinoma and Nasopharyngeal inflammation was detected by Immunohistochemistry. The association between the different degrees of differentiation ages, TNM clinical stages was evaluated. Collect medical records of patients diagnosed as 60 cases of nasopharyngeal carcinoma, chronic inflammation of patient records 24 cases of nasopharyngeal, its corresponding paraffin-embedded tissue, with each successive slice thickness 4μm. Making tissue microarray, use 4h2 mouse anti-human prion protein monoclonal antibody as an anti-1 provided by the Wuhan Institute of Virology, Chinese Academy of virology laboratory Chaoyang- Li. Related kit was purchased from Beijing Zhongshan Golden Bridge Biotechnology. Correlation detection Pr Pc in different tissues and expression analysis and clinical parameters between patients with nasopharyngeal carcinoma. Results: Pr Pc expression of inflammation in the nasopharynx, nasopharyngeal carcinoma : Pr Pc inflammation in the nasopharynx were positive, the positive rate was 8.3%( 2/24), only two cases, In nasopharyngeal carcinoma mainly localized in the cytoplasm and the cytoplasm, the positive rate was 30%( 18/60) was significantly higher than the nasopharynx inflammatory tissues(P <0.05). The expression of the relationship between the clinical parameters associated prion protein :(1) Pr Pc in nasopharyngeal carcinoma high grade, poorly differentiated positive expression rates of 7.69%( 1/13) 36.17%( 17/47) respectively, and the difference was statistically significant(P <0.05).( 2) In the 43 cases with lymph node metastasis of clinical data, there are 13 cases of positive expression, 12 cases without lymph node metastasis in 5 cases positive, lymph node metastasis and whether positive no significant(P> 0.05).(3) TNM staging : 20 cases I, Ⅱ, 16 cases, 15 cases of stage Ⅲ, stage Ⅳ 9 cases, prion protein decreased intensity with TNM stage and higher grade, four overall difference has significant correlation(P < 0.05); Prion protein I, Ⅱ period of progressive enhancement expression was statistically significant(P <0.05); while I + Ⅱ and Ⅲ + Ⅳ stage of expression differences was statistically significant(P <0.05). Condusions: In a high state of nasopharyngeal carcinoma Pr Pc expression is likely to participate in local invasion and metastasis of nasopharyngeal carcinoma tumor tissue and molecular pathways of distant metastasis, and bear important physiological functions in the process of tumor cells, and Pr Pc expression and TNM stage, predicting distant metastasis also has important clinical significance. Local control and regional control is poor, is one of the important causes of distant metastasis. T in patients with late stage tumors near the brain, severely limits the angle and dose of radiation therapy, radiation therapy can not make effective, leading to a local regional control rate is not ideal. It has been confirmed is closely related to the development of EB virus infection and nasopharyngeal carcinoma, the expression of Pr Pc in NPC cells is affected by EB virus infection needs further study, obviously Pr Pc expression in malignant tumors, which in tumor cells whether he will take the anti-apoptotic effect and is widely present in the cytoplasm of whether to undertake certain functions of tumor cell growth, pending further study.
